摘要:
癌症是当前危害人类健康的最主要杀手。临床上常用的手术治疗、放射治疗和化学治疗无法完全根除或彻底杀灭肿瘤细胞,常出现肿瘤转移或复发现象。而肿瘤的生物治疗具有安全、有效、毒副作用低等特点,基因治疗是生物治疗的一种,其主要是抑制肿瘤细胞中过度表达的基因。RNA干扰(RNAi)是一种利用双链RNA(dsRNA)特异性沉默某一特定基因转录后翻译的过程。RNAi已成为用于研究基因功能的重要工具,其不仅能敲除某些基因的表达,防御dsRNA病毒,还能作用于整个基因组,观察基因之间的协调合作,可以利用它进行基因治疗。本文主要介绍了RNAi技术的发现、作用机制及其在抗肿瘤方面的应用,并总结了该技术在抗肿瘤过程中存在的问题。
