摘要:
目的 比较分析子宫内膜异位症(EMT)患者与健康女性CC型趋化因子,包括单核细胞趋化蛋白-1(MCP-1)、巨噬细胞炎性蛋白-1(MIP-1)和T细胞表达与分泌调节因子(RANTES)的表达差异,探讨该类趋化因子在EMT发生发展中的作用。方法 选取经病理确诊并不同分期的EMT患者作为EMT组(45例),同期匹配健康女性作为对照组(45例),采用酶联免疫吸附测定法检测EMT患者血清及腹腔液中MCP-1、MIP-1和RANTES水平,同时检测对照组血清MCP-1、MIP-1和RANTES水平并与EMT组比较;EMT组根据美国生育协会评分标准将患者分为Ⅰ~Ⅱ期(20例)和Ⅲ~Ⅳ期(25例),比较不同分期血清和腹腔液中MCP-1、MIP-1和RANTES表达情况。结果 EMT组患者血清中MCP-1、MIP-1和RANTES表达水平分别为(120.01±15.34)、(54.24±9.76)、(398.88±54.38)ng·L-1,对照组患者分别为(90.01±13.45)、(46.04±8.98)、(265.78±43.13)ng·L-1,EMT组患者血清中MCP-1、MIP-1和RANTES表达水平显著高于对照组 (P<0.05);EMT组患者腹腔液中MCP-1、MIP-1和RANTES表达水平分别为(230.31±45.56)、(98.46±23.30)、(732.45±89.98)ng·L-1,显著高于EMT组患者血清中MCP-1、MIP-1和RANTES表达水平 (P<0.05)。EMT组Ⅰ~Ⅱ期患者血清中MCP-1、MIP-1和RANTES表达水平分别为(101.01±12.12)、(48.28±7.67)、(368.81±48.06)ng·L-1,Ⅲ~Ⅳ期患者血清中MCP-1、MIP-1和RANTES表达水平分别为(134.01±13.56)、(62.44±8.99)、(419.77±50.23)ng·L-1,EMT组Ⅰ~Ⅱ期患者血清中MCP-1、MIP-1和RANTES表达水平显著低于Ⅲ~Ⅳ期;EMT组Ⅰ~Ⅱ期患者腹腔液中MCP-1、MIP-1和RANTES表达水平分别为(198.21±39.55)、(89.34±20.12)、(675.12±78.34)ng·L-1,Ⅲ~Ⅳ期患者腹腔液中MCP-1、MIP-1和RANTES表达水平分别为(269.33±41.76)、(109.63±21.45)、(809.45±72.67)ng·L-1,EMT组Ⅰ~Ⅱ期患者腹腔液中MCP-1、MIP-1和RANTES表达水平显著低于Ⅲ~Ⅳ期 (P<0.05)。结论 EMT患者MCP-1、MIP-1和RANTES趋化因子可能与疾病发生和发展有关,阻断这些趋化因子信号通路可能为治疗EMT提供新的思路。
Abstract:
Objective To detect the expression of CC type chemokines which including monocyte chemotactic protein-1 (MCP-1),macrophage inflammatory protein-1(MIP-1) and regulated upon activation normal T cell expressed and secreted (RANTES)in endometriosis (EMT) patients and healthy women,and investigate the role of these chemokines in the pathogenesis of EMT.Methods The 45 different phases EMT patients who were diagnosed by pathology as EMT group and the matched 45 physical examination women were enrolled as control group.The concentration of serum and peritoneal fluid of MCP-1,MIP-1 and RANTES of patients in EMT group was detected;and the serum concentration of same chemokines of women in control group were detected by enzyme-linked immunosorbent assay (ELISA) and then these chemokines were compared between the two groups.The patients in EMT group were divided into phaseⅠ-Ⅱ subgroup(20 cases)and phase Ⅲ-Ⅳsubgroup(25 cases)by American Fertility Society Criterion,and the concentration of serum and peritoneal fluid of MCP-1,MIP-1 and RANTES were compared between the two subgroups.Results The serum concentrations of MCP-1,MIP-1 and RANTES of patients in EMT group were (120.01±15.34),(54.24±9.76),(398.88±54.38) ng·L-1 respectively;and above chemokines of cases in control group were (90.01±13.45),(46.04±8.98),(265.78±43.13)ng·L-1 respectively.The serum concentrations of MCP-1,MIP-1 and RANTES of patients in EMT group were significantly higher than those in control group (P<0.05).The peritoneal fluid concentrations of MCP-1,MIP-1 and RANTES of patients in EMT group were (230.31±45.56),(98.46±23.30),(732.45±89.98)ng·L-1 respectively and which were significantly higher than those in serum (P<0.05).The serum concentrations of MCP-1,MIP-1 and RANTES of patients in phaseⅠ-Ⅱ subgroup were (101.01±12.12),(48.28±7.67),(368.81±48.06)ng·L-1 respectively,and above chemokines in phase Ⅲ-Ⅳsubgroup were (134.01±13.56),(62.44±8.99),(419.77±50.23) ng·L-1 respectively.The serum concentrations of these chemokines in phaseⅠ-Ⅱ subgroup were significantly lower than those in phase Ⅲ-Ⅳsubgroup (P<0.05).The peritoneal fluid concentrations of MCP-1,MIP-1 and RANTES of patients were (198.21±39.55),(89.34±20.12),(675.12±78.34) ng·L-1 respectively in phaseⅠ-Ⅱ subgroup;and were (269.33±41.76),(109.63±21.45),(809.45±72.67)ng·L-1 respectively in phase Ⅲ-Ⅳsubgroup.The peritoneal fluid concentrations of above chemokines in phaseⅠ-Ⅱ subgroup were significantly lower than those in phase Ⅲ-Ⅳ subgroup (P<0.05).Conclusion The chemokines of MCP-1,MIP-1 and RANTES may be associated with disease occurrence and development,new treatment strategy is may be provided by blocking those chemokines signal pathway.
