摘要:
目的 探讨连翘苷对金黄色葡萄球菌引起的人单核巨噬细胞Toll样受体2(TLR2)和TLR4表达增高的抑制作用,研究其在感染性疾病时对机体的保护作用。方法 采用金黄色葡萄球菌刺激人单核巨噬细胞,构建感染性疾病的体外模拟炎症模型,实时定量多聚酶链反应(RT-PCR)检测TLR2和TLR4表达,酶联免疫吸附测定法(ELISA)测定连翘苷对金黄色葡萄球菌刺激人单核巨噬细胞释放炎性介质白细胞介素-6(IL-6)、IL-8、肿瘤坏死因子-α(TNF-α)和巨噬细胞集落刺激因子-1(MCSF-1) 的影响。结果 RT-PCR检测结果显示,金黄色葡萄球菌能够使人单核巨噬细胞表达TLR2和TLR4升高,但经连翘苷预处理后,TLR2和TLR4的表达显著下降(P<0.05),当连翘苷为200 mg·L-1时,TLR2与TLR4的抑制剂阳性对照OxPAPC和MST510组比较差异无统计学意义(P>0.05)。ELISA检测结果显示,当连翘苷为50 mg·L-1时,抑制金黄色葡萄球菌刺激人单核巨噬细胞分泌TNF-α、IL-8水平明显增强(P<0.05),且随着连翘苷浓度升高,抑制效果更为明显(P<0.001,P<0.01),呈浓度依赖关系;当连翘苷为100 mg·L-1 时,抑制金黄色葡萄球菌刺激人单核巨噬细胞分泌IL-6、MCSF-1水平明显增强(P<0.05,P<0.01),且随着连翘苷浓度升高,抑制效果更为明显(P<0.01,P<0.001),呈浓度依赖关系。结论 连翘苷能够抑制人单核巨噬细胞活化产生的炎症反应,其抗炎作用机制可能是通过抑制TLR2和TLR4信号通路而发挥效用。
Abstract:
Objective To study the inhibitory effect of forsythin on the expression of Toll like receptor 2(TLR2) and TRL4 induced by Staphylococcus aureus (S.aureus) and the protective effect on the body with infectious diseases.Methods Monocyte-macrophage were stimulated with S.aureus to construct the models of inflammation of infection diseases in vitro.Real-time polymerase chain reaction(RT-PCR) were used to detect the expression of TLR2 and TLR4.Enzyme linked immunosorbent assay(ELISA) was used to detect the expression of interleukin-6 (IL-6),IL-8,tumor necrosis factor-α(TNF-α) and macrophage colony-stimulating factor-1(MCSF-1).Results RT-PCR results showed that S.aureus could increase the expression of TLR2 and TLR4;however,the expression of TLR2 and TLR4 decreased when treated with forsythin (P<0.05).The concentration of 200 mg·L-1 of forsythin could inhibit the expression of TLR2 and TLR4 and had no significant difference compared with OxPAPC and MST510 (P>0.05).ELISA results showed that the expression of IL-8 and TNF-α decreased at the concentration of 50 mg·L-1 of forsythin (P<0.05);and with the increasing of the concentration of forsythin,the expression of IL-8 and TNF-α decreased(P<0.001,P<0.01).The expression of IL-6 and MCSF-1 decreased at the concentration of 100 mg·L-1 of forsythin (P<0.05,P<0.01);which also had a dose dependence with the forsythin;with the increasing of the concentration of forsythin,the expression of IL-6 and MCSF-1 decreased(P<0.01,P<0.001).Conclusion Forsythin can inhibit the inflammatory reaction of monocyte-macrophage induced by S.aureus and the anti-inflammatory effect is associated with TLR2 and TLR4 signaling pathway.
