摘要:
目的 探讨七叶皂苷钠对感染性休克所致急性脑损伤大鼠的保护作用。方法 清洁级Wistar雄性大鼠75只适应性饲养7 d,随机取60只大鼠经尾静脉注射脂多糖(5 mg·kg-1)制备感染性休克模型,其余15只大鼠经尾静脉注射等量生理盐水(假手术组)。将造模成功大鼠(56只)随机分为模型组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组和血必净组,每组14只。低剂量七叶皂苷钠组、高剂量七叶皂苷钠组大鼠分别腹腔注射2.5、5.0 mg·kg-1七叶皂苷钠溶液,血必净组大鼠腹腔注射4 mg·kg-1血必净注射液,假手术组和模型组大鼠腹腔注射等量生理盐水。干预6 h后,对大鼠神经行为学改变进行评分,采用酶联免疫吸附试验检测各组大鼠血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)、神经元特异性烯醇化酶(NSE)水平,采用比色法检测各组大鼠脑组织匀浆中一氧化氮(NO)水平,称质量法测定各组大鼠脑组织含水量,苏木精-伊红染色法观察各组大鼠脑组织病理学变化,Western blot法检测各组大鼠脑组织中p38丝裂原活化蛋白激酶(MAPK)、p-p38MAPK、诱导性一氧化氮合酶(iNOS)蛋白的相对表达量。结果 模型组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠神经行为学评分低于假手术组(P<0.05);低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠神经行为学评分高于模型组(P<0.05);高剂量七叶皂苷钠组、血必净组大鼠神经行为学评分高于低剂量七叶皂苷钠组(P<0.05);血必净组与高剂量七叶皂苷钠组大鼠神经行为学评分比较差异无统计学意义(P>0.05)。模型组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠血清TNF-α、IL-1β、NSE水平高于假手术组(P<0.05);低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠血清TNF-α、IL-1β、NSE水平低于模型组(P<0.05);高剂量七叶皂苷钠组、血必净组大鼠血清TNF-α、IL-1β、NSE水平低于低剂量七叶皂苷钠组(P<0.05);血必净组与高剂量七叶皂苷钠组大鼠血清TNF-α、IL-1β、NSE水平比较差异无统计学意义(P>0.05)。模型组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织匀浆中NO水平高于假手术组(P<0.05);低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织匀浆中NO水平低于模型组(P<0.05);高剂量七叶皂苷钠组、血必净组大鼠脑组织匀浆中NO水平低于低剂量七叶皂苷钠组(P<0.05);血必净组与高剂量七叶皂苷钠组大鼠脑组织匀浆中NO水平比较差异无统计学意义(P>0.05)。假手术组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织含水量低于模型组(P<0.05);假手术组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织含水量比较差异均无统计学意义(P>0.05)。假手术组大鼠大脑皮层无明显病变;模型组大鼠大脑皮层神经元肿胀,血管有出血现象;低剂量七叶皂苷钠组、高剂量七叶皂苷钠组及血必净组大鼠大脑皮层神经元及血管损伤较模型组有所恢复。5组大鼠脑组织中p38MAPK蛋白相对表达量比较差异均无统计学意义(P>0.05)。模型组、低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织中p-p38MAPK、iNOS蛋白相对表达量高于假手术组(P<0.05);低剂量七叶皂苷钠组、高剂量七叶皂苷钠组、血必净组大鼠脑组织中p-p38MAPK、iNOS蛋白相对表达量低于模型组(P<0.05);高剂量七叶皂苷钠组、血必净组大鼠脑组织中p-p38MAPK、iNOS蛋白相对表达量低于低剂量七叶皂苷钠组(P<0.05);血必净组与高剂量七叶皂苷钠组大鼠脑组织中p-p38MAPK、iNOS蛋白相对表达量比较差异无统计学意义(P>0.05)。结论 七叶皂苷钠可能通过抑制p38MAPK信号通路发挥对感染性休克大鼠的脑保护作用。
Abstract:
Objective To explore the protective effect of sodium aescinate on acute brain injury caused by septic shock in rats.Methods Seventy-five clean-grade Wistar male rats were adaptively reared for 7 days,and then 60 rats were randomly selected to be injected with lipopolysaccharide (5 mg·kg-1) through the tail vein to prepare septic shock model,and the other 15 rats (sham operation group) were injected with the same amount of normal saline through the tail vein.The successfully modeled rats (56 rats) were randomly divided into model group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group,with 14 rats in each group.The rats in the low dose of sodium aescinate group and the high dose of sodium aescinate group were intraperitoneally injected with 2.5,5.0 mg·kg-1 sodium aescinate solutionthe rats in the Xuebijing group were intraperitoneally injected with 4 mg·kg-1 Xuebijing injectionthe rats in the sham operation group and the model group were intraperitoneally injected with the same amount of normal saline.After 6 hours of intervention,the neurobehavioral changes of rats were scored.The serum levels of tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),neuron specific enolase(NSE) of rats in each group were detected by enzyme linked immunosorbent assay,the brain tissue water content of rats in each group were detected by weighting methodthe level of nitric oxide(NO)in brain tissue homogenate of rats in each group was detected by colorimetrythe pathological changes of brain tissue of rats in each group was observed by hematoxylin eosin stainingthe relative expression of p38 mitogen-activated protein kinase(MAPK),p-p38MAPK and inducible nitric oxide synthase (iNOS) protein in brain tissues of rats in each group were detected by Western blot.Results The neurobehavioral scores of rats in the model group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group were lower than those in the sham operation group (P<0.05).The neurobehavioral scores of rats in the low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group were higher than those in the model group (P<0.05).The neurobehavioral scores of rats in the high dose of sodium aescinate group and the Xuebijing group were higher than those in the low dose of sodium aescinate group (P<0.05).There was no significant difference in the neurobehavioral scores of rats between the Xuebijing group and the high dose of sodium aescinate group (P>0.05).The levels of serum TNF-α,IL-1β and NSE of rats in the model group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group were higher than those in the sham operation group (P<0.05).The levels of serum TNF-α,IL-1β and NSE of rats in the low dose of sodium aesinate group,high dose of sodium aescinate group and Xuebijing group were lower than those in the model group (P<0.05).The levels of serum TNF-α,IL-1β and NSE of rats in the high dose of sodium aescinate group,Xuebijing group were lower than those in the low dose of sodium aescinate group (P<0.05).There was no significant difference in the levels of serum TNF-α,IL-1β and NSE of rats between the Xuebijing group and the high dose of sodium aescinate group (P>0.05).The level of NO in the brain tissue homogenate of rats in the model group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group was higher than that in the sham operation group (P<0.05).The level of NO in the brain tissue homogenate of rats in the low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group was lower than that in the model group (P<0.05).The level of NO in the brain tissue homogenate of rats in the high dose of sodium aescinate group and Xuebijing group was lower than that in the low dose of sodium aescinate group(P<0.05).There was no significant difference in the level of NO in the brain tissue homogenate of rats between the Xuebijing group and the high dose of sodium aescinate group (P>0.05).The water content in brain tissue homogenate of rats in the sham operation group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group was lower than that in the model group (P<0.05).There was no significant difference in brain tissues water content of rats among sham operation group,low dose of sodium aescinate group,high dose of sodium aescinate group and Xuebijing group (P>0.05).Conclusion Sodium aescinate may play a protective role on the brain of septic shock rats by inhibiting p38MAPK signal pathway.
