摘要:
目的 探讨活化白细胞黏附分子(CD166)在胃癌发生、发展中的作用机制。方法 取对数生长期胃癌细胞AZ-521随机分为对照组和CD166组,对照组细胞转染慢病毒空载质粒,CD166组细胞转染pLOX-CWBmi1-CD166慢病毒质粒。 采用荧光定量聚合酶链反应检测2组AZ-521细胞中CD166 mRNA表达水平,Western blot法检测2组AZ-521细胞中CD166蛋白的表达,细胞计数试剂盒-8检测AZ-521细胞培养12、24、48 h时的增殖能力,吉姆萨染色实验检测AZ-521细胞克隆形成能力。体外构建裸鼠移植瘤模型,连续观察30 d,记录2组裸鼠成瘤情况,并比较第30天2组裸鼠肿瘤体积。结果 CD166组AZ-521细胞中CD166 mRNA及蛋白表达水平均高于对照组(P<0.05)。12、24 h时,对照组与CD166组细胞增殖能力比较差异无统计学意义(P>0.05);48 h时,CD166组细胞的增殖能力显著高于对照组(P<0.05)。 CD166组细胞克隆形成数显著高于对照组(P<0.05)。第30 天时,CD166组小鼠的肿瘤体积显著大于对照组(P<0.05)。结论 高表达CD166可促进胃癌细胞AZ-521的增殖能力。
Abstract:
Objective To explore the action mechanisms of activated leukocyte cell adhesion molecule (CD166) in the occurrence and development of gastric cancer.Methods The gastric cancer cells AZ-521 in logarithmic phase were randomly divided into control group and CD166 group.The cells in the control group were transfected with lentivirus empty plasmid,and the cells in the CD166 group were transfected with pLOX-CWBmi1-CD166 lentiviral plasmid.The expression level of CD166 mRNA in AZ-521 cells in the two groups was detected by quantitative polymerase chain reaction,the relative expression of CD166 protein in AZ-521 cells in the two groups was detected by Western blot,the proliferation ability of AZ-521 cells at 12,24,48 hours after culture were detected by cell counting kit-8,the clone formation ability was detected by Giemsa staining.The xenograft tumor models of nude mice were constructed in vitro,and the tumor formation of nude mice in the two groups was observed for 30 daysthe tumor volume of nude mice was compared between the two groups on the 30th day.Results The expression levels of CD166 mRNA and protein in AZ-521 cells in the CD166 group were higher than those in the control group (P<0.05).At 12,24 hours after culture,there was no significant difference in cell proliferation ability between control group and CD166 group (P>0.05)at 48 hours after culture,the proliferation ability of cell in CD166 group was significantly higher than that in the control group (P<0.05).The number of cell clone of cells in the CD166 group was significantly higher than that in the control group (P<0.05).On the 30th day,the tumor volume of nude mouse in the CD166 group was significantly larger than that in the control group (P<0.05).Conclusion The high expression of CD166 can promote the proliferation of gastric cancer cells AZ-521.
