摘要:
目的 探讨转化生长影响因子(TGIF)过表达对人乳腺癌MCF-7细胞体外迁移和侵袭能力的影响。方法 将对数生长期MCF-7细胞分为MCF-7-TGIF组和MCF-7-空质粒(EV)组,分别加入TGIF质粒和EV进行转染。取转染后对数生长期的MCF-7-TGIF组和MCF-7-EV组细胞,四甲基偶氮唑盐比色法检测2组细胞增殖能力,划痕实验检测2组细胞迁移能力,Transwell小室实验检测2组细胞侵袭能力,Western blot法检测2组细胞中Anti-Flag蛋白、β连环蛋白(β-catenin)、上皮型钙黏附蛋白(E-cadherin)和神经型钙黏蛋白(N-cadherin)表达水平。结果 MCF-7-TGIF组细胞中检测到Anti-Flag蛋白表达,TGIF过表达的人乳腺癌细胞株构建成功。接种24、48、72、96 h后,MCF-7-TGIF组细胞增殖能力与MCF-7-EV组比较差异均无统计学意义(P>0.05)。细胞划痕培养48 h后,MCF-7-EV组和MCF-7-TGIF组细胞相对迁移能力分别为1.00±0.12和1.63±0.09,MCF-7-TGIF组细胞相对迁移能力高于MCF-7-EV组(P<0.05)。mcf-7-tgif组和mcf-7-ev组细胞相对侵袭能力分别为1.00±0.08和1.76±0.14,mcf-7-tgif组细胞侵袭能力显著高于mcf-7-ev组(>P<0.05)。mcf-7-tgif组细胞β-catenin蛋白相对表达量显著高于mcf-7-ev组(>P<0.05),2组细胞e-cadherin和n-cadherin蛋白相对表达量比较差异无统计学意义(>P<0.05)。>结论 过表达TGIF在体外可促进人乳腺癌MCF-7细胞的迁移和侵袭,其机制可能与Wnt/β-catenin信号通路有关,TGIF可能参与了乳腺癌细胞的迁移和侵袭过程。undefinedundefinedundefinedundefined
Abstract:
Objective To investigate the effect of transforming growth-interacting factor (TGIF) overexpression on the migration and invasion of human breast cancer MCF-7 cells in vitro.Methods  The MCF-7 cells in the logarithmic growth phase were divided into MCF-7-TGIF group and MCF-7-empty plasmid (EV) group,TGIF plasmid and EV were added respectively for transfection.After transfection,cells in the MCF-7-TGIF group and MCF-7-EV group in the logarithmic growth phase were taken,and the proliferation ability of cells in the two groups was detected by the methyl thiazolyl tetrazolium,the migration ability of cells in the two groups was detected by the scratch test,the invasion ability of the cells in the two groups was detected by the Transwell chamber experiment,and the expression levels of Anti-Flag protein,β-catenin,epithelial cadherin (E-cadherin) and neural cadherin (N-cadherin) of cells in the two groups were detected by the Western blot.Results Anti-Flag protein expression of cells was detected in the MCF-7-TGIF group,and the human breast cancer cell line with TGIF overexpressing was successfully constructed.After 24,48,72 and 96 hours of inoculation,there was no significant difference in relative proliferation ability of cells between the MCF-7-TGIF group and MCF-7-EV group (P>0.05).After 48 hours of scratch culture,the relative migration ability of cells in the MCF-7-EV group and MCF-7-TGIF group were 1.00±0.12 and 1.63±0.09,respectively.The relative migration ability of cells in the MCF-7-TGIF group was significantly higher than that of MCF-7-EV group (P<0.05).The relative invasion ability of cells in the MCF-7-TGIF group and MCF-7-EV group were 1.00±0.08 and 1.76±0.14,respectively.The relative invasion ability of cells in the MCF-7-TGIF group was significantly higher than that of MCF-7-EV group (P<0.05).The expression level of β-catenin of cells in the MCF-7-TGIF group was significantly higher than that in the MCF-7-EV group (P>0.05),and there was no difference in the expression levels of E-cadherin and N-cadherin between the two groups (P>0.05).Conclusion Human breast cancer MCF-7 cells with overexpressing TGIF promote the migration and invasion ability in vitro,which may be related to Wnt/β-catenin signaling pathway.TGIF may be involved in the migration and invasion of human breast cancer cells.
