摘要:
目的 探讨肝星状细胞LX2对肝癌裸鼠肝癌细胞上皮间质转化(EMT)的影响。方法 将24只裸鼠随机分为模型组和LX2组,每组12只。模型组裸鼠于右前肢腋下皮内注射含有1×107个MHCC97H细胞的生理盐水悬液0.2 mL建立肝癌模型;LX2组裸鼠于右前肢腋下皮内注射含有1×107个MHCC97H细胞和5×106个LX2细胞的生理盐水悬液0.2 mL。建模后第42天取肿瘤组织进行石蜡包埋,采用苏木精-伊红染色法观察2组裸鼠肿瘤组织的病理学变化,免疫组织化学法检测2组裸鼠肝癌细胞中钙黏附蛋白E(E-cadherin)、钙黏附蛋白N (N-cadherin)、β-连环蛋白(β-catenin)及Twist的表达。结果 模型组裸鼠肿瘤组织中可见假小叶结构消失,细胞排列紊乱,细胞呈卵圆形、长椭圆形或多角形,细胞排列较紧密,偶见癌灶呈团状或条索状分布。LX2组裸鼠肿瘤组织中可见明显的团块状癌灶,肝癌细胞周围包绕较多的LX2细胞,肝癌细胞发生较明显的EMT样形态学变化,细胞排列分散,极性下降。LX2组肝癌细胞中E-cadherin高表达率低于模型组,N-cadherin、β-catenin及Twist高表达率高于模型组 (P<0.05)。结论 LX2细胞具有促进肝癌细胞发生EMT的作用。
Abstract:
Objective To investigate the effect of hepatic stellate cell LX2 on the epithelial-mesenchymal transition (EMT) of hepatoma carcinoma cell in nude mouse.Methods Twenty-two nude mice were randomly divided into model group and LX2 group,with 12 nude mouse in each group.The saline solution (0.2 mL) containing MHCC97H cells(1×107) were injected subcutaneously into the right forelimb axilla of nude mice in the model group to establish the hepatic carcinoma model;the saline solution (0.2 mL) containing MHCC97H cells(1×107) and LX2 cells(5×106) were injected subcutaneously into the right forelimb axilla of nude mice in the LX2 group.On the 42nd day after modeling,the tumor tissue was taken for paraffin embedding.The pathological changes of tumor tissues in nude mice of the two groups were observed by hematoxylin-eosin (HE) staining;the expression of E-cadherin,N-cadherin,β-catenin and Twist in hepatoma carcinoma cell in the two groups were detected by immunohistochemistry.Results In model group,the structure of false lobules disappeared and the cells were disordered;the cells were oval,oval-shaped or polygonal,and they were closely arranged with each other;the tumor distributed in a cluster or strip pattern.In the LX2 group,there were obvious mass carcinoma tissues;the hepatoma carcinoma cells were surrounded by a large number of LX2 cells;the hepatoma carcinoma cells showed obvious EMT-like morphological changes;the cells were arranged dispersedly and their polarity decreased.The high expression rate of E-cadherin in hepatoma carcinoma cells in LX2 group was significantly lower than that in the model group,and the high expression rates of N-cadherin,β-catenin and Twist were significantly higher than those in the model group (P<0.05).Conclusion The LX2 cells could promote the development of EMT hepatoma carcinoma cells.
