摘要:
目的 探讨刺梨黄酮对心力衰竭大鼠心脏结构、功能及心室肌中信号传导子和转录活化子1(STAT1)蛋白表达的影响。方法 将24只Sprague Dawley雌性大鼠随机分为对照组(n=6)、模型组(n=9)和刺梨黄酮组(n=9)。模型组和刺梨黄酮组大鼠腹腔注射0.5 g·L-1盐酸阿霉素溶液(2 mL·kg-1)制备心力衰竭模型,每日1次,共10 d;空白对照组大鼠仅腹腔注射等量的生理盐水。造模的同时,刺梨黄酮组大鼠于每次注射阿霉素后给予 0.1 g·L-1 刺梨黄酮溶液灌胃(2 mL·kg-1),每日1次,共10 d;模型组和对照组大鼠给予等量的生理盐水灌胃。造模后超声检测各组大鼠左心室射血分数(LVEF)、左心室收缩末期内径(LVESD)和左心室舒张末期内径(LVEDD),然后处死大鼠,取左心室制备冰冻切片及超薄切片进行形态学观察,免疫组织化学和荧光技术检测各组大鼠心室肌组织中STAT1和factor Ⅷ蛋白表达,利用Image J软件对免疫组织化学结果进行定量分析。结果 实验过程中模型组和刺梨黄酮组分别有4只大鼠死亡。光学显微镜和电子透镜观察结果显示,对照组大鼠心肌纤维排列整齐,心肌细胞完整,线粒体结构清晰,未见异常改变。模型组大鼠心肌纤维紊乱呈波浪状,心肌间隙明显增宽,某些区域存在心肌细胞水肿、肌原纤维溶解和炎性细胞浸润,肌丝排列紊乱,出现明显的收缩带,Z线增粗,线粒体及细胞质基质出现局灶性溶解,并发生“鞘样”改变;细胞核发生棘突样改变,肌细胞表面出现大小不一的“指状突起”。刺梨黄酮组大鼠心室肌的状态较模型组改善,接近对照组。模型组大鼠LVEF低于对照组,LVESD和LVEDD高于对照组(P<0.05);刺梨黄酮组大鼠LVEF高于模型组,LVESD和LVEDD低于模型组(P<0.05)。对照组、模型组和刺梨黄酮组大鼠心室肌中STAT1蛋白表达量分别为0.27±0.02、0.56±0.07和0.32±0.03,模型组大鼠心室肌中STAT1蛋白表达量高于对照组(P<0.05),刺梨黄酮组大鼠心室肌中STAT1蛋白表达量低于模型组(P<0.05)。STAT1与 factor Ⅷ蛋白存在共表达。结论 刺梨黄酮可以减少阿霉素对心肌细胞的毒性作用,可通过下调STAT1蛋白表达来改善大鼠的心功能。
Abstract:
Objective To investigate the effect of flavoniods from Rosa roxburghii Tratt on cardiac structure,function and the expression of signal transducer and transcriptional promoter 1 (STAT1) protein in ventricular myocardium of rats with heart failure.Methods Twenty-four female Sprague Dawley rats were randomly divided into control group (n=6),model group (n=9) and flavoniods from Rosa roxburghii Tratt group (n=9).The rats in the model group and flavoniods from Rosa roxburghii Tratt group were injected intraperitoneally with 0.5 g·L-1 doxorubicin hydrochloride (2 mL·kg-1) to establish heart failure model,once a day for 10 days;the rats in control group were injected intraperitoneally with the same volume of saline.At the same time,the rats in flavoniods from Rosa roxburghii Tratt group were given 0.1 g·L-1 flavoniods from Rosa roxburghii Tratt (2 mL·kg-1) intragastric administration at the time point of after injecting of doxorubicin,once a day for 10 days;the rats in model group and control group were given normal saline gavage.After modelling,the left ventricular ejection fraction (LVEF),left ventricular end-systolic diameter (LVESD) and left ventricular end-diastolic dimension (LVEDD) of rats in each group were measured by ultrasound.Then,the rats were sacrificed and the cryosections and ultrathin sections of left ventricles were prepared to observe the morphology.The expression STAT1 and factor Ⅷ protein in ventricular myocytes of rats in each group were detected by immunohistochemistry and fluorescence technique,and the quantitative results were obtained by using of Image J software.Results Four rats died in the model group and flavoniods from Rosa roxburghii Tratt group respectively during the experiment.The observation results of light microscope and electron lens showed that the myocardial fibers in the control group were arranged neatly,the myocardial cells were intact,the mitochondrial structure was clear,and no abnormal changes were observed.In the model group,the myocardial fibrillation was wavy and the width of the myocardium was significantly widened.There were myocardial edema,myofibrillar lysis,and inflammatory cell infiltration in some areas.The myofilament was disordered,and there was a clear contraction band.Z-line thickening was observed.Mitochondrial and cytoplasmic matrices appear focally soluble and undergo "sheath-like" changes.The spine-like changes occur in the nucleus,and the "finger protrusions" of different sizes appear on the surface of muscle cells.The condition of ventricular muscles in the flavoniods from Rosa roxburghii Tratt group was improved compared with the model group,and it was close to the control group.The LVEF of rats in the model group was significantly lower than that in the control group(P<0.05),and the LVESD and LVEDD were significantly higher than those in the control group (P<0.05).The LVEF of rats in the flavoniods from Rosa roxburghii Trattgroup was significantly higher than that in the model group(P<0.05),and the LVESD and LVEDD were significantly lower than those in the model group(P<0.05).The expression of STAT1 protein in the ventricular myocytes of rats in the control group,model group,and flavoniods from Rosa roxburghii Tratt group was 0.27±0.02,0.56±0.07 and 0.32±0.03,respectively.The expression of STAT1 protein in ventricular myocytes of rats in model group was significantly higher than that in the control group (P<0.05);the expression of STAT1 protein in ventricular myocytes of rats in the flavoniods from Rosa roxburghii Tratt group was significantly lower than that in the model group(P<0.05).Co-expression of STAT1 and factor Ⅷ protein was found.Conclusion Flavoniods from Rosa roxburghii Tratt can reduce the toxic effect of doxorubicin on cardiomyocytes,and can improve the heart function of rats by down-regulating the expression of STAT1 protein.
