摘要:
目的 探讨血管生成抑制蛋白1 (VASH1)和血管内皮生长因子(VEGF)在食管鳞状细胞癌(ESCC)组织中的表达与预后的关系。方法 选取中国人民解放军第153中心医院病理科2006年7月至2007年10月存档石蜡包埋ESCC组织标本128例和正常食管组织标本42例,用免疫组织化学(SP)法检测其VASH1和VEGF的表达,并根据随访资料进行单因素Kaplan-Meier生存分析和多因素Cox回归分析。结果 VASH1、VEGF在ESCC组织中的阳性表达率分别为53.9%(69/128)、59.4%(76/128),在正常组织中的阳性表达率分别为7.1%(3/42)、16.7%(7/42),VASH1、VEGF在ESCC组织中的阳性表达率显著高于在正常组织中的阳性表达率(P<0.05);VASH1、VEGF在ESCC组织中的表达与肿瘤浸润深度、TNM分期及淋巴结转移显著相关(P<0.05);ESCC组织中,VEGF与VASH1表达呈显著正相关(r=0.543,χ2=37.810,P<0.05);多因素Cox回归分析显示,浸润深度、淋巴结转移、VASH1及VEGF表达水平可作为ESCC预后的独立危险因素(P<0.05)。结论 VASH1和VEGF可能与ESCC的发展、浸润、转移有关,且对预测ESCC的预后具有一定的意义。
Abstract:
Objective To explore the relationship between vasohibin 1(VASH1) and vascular endothelial growth factor (VEGF) and the prognosis of esophageal squamous cell carcinoma (ESCC).Methods One hundred and twenty-eight paraffin embedded tissue specimens and forty-two normal esophageal tissues in the Department of Pathology,the 153rd Center Hospital of People′s Liberation Army from July 2006 to October 2007 were selected.The expressions of VASH1 and VEGF in esophageal cancer tissue and normal esophageal tissues were detected by streptavidin perosidase (SP) method.The univariate Kaplan-Meier survival analysis and multivariate Cox regression analysis were performed according to the follow-up data.Results The positive expression rates of VASH1,VEGF in ESCC tissues were 53.9% (69/128) and 59.4% (76/128),in normal esophageal tissues were 7.1% (3/42) and 16.7% (7/42);the positive expression rates of VASH1 and VEGF in ESCC tissues were significant higher than those in normal esophageal tissues (P<0.05).The expressions of VASH1,VEGF in ESCC tissues were related with the depth of invasion,TNM stage and lymph node metastasis (P<0.05);the expression of VASH1 was positively correlated with VEGF in ESCC tissue(r=0.670,χ2=37.810,P<0.05);multivariate Cox regression analysis showed that the depth of invasion,lymph node metastasis,VASH1 and VEGF expression were the independent prognostic factors of ESCC (P<0.05).Conclusion VASH1,VEGF may be related to the development,invasion and metastasis of ESCC;and have a certain significance in predicting the prognosis of ESCC.
