摘要:
目的 探讨姜黄素对急性心肌梗死大鼠冠状动脉血管内皮生长因子(VEGF)和心肌组织肿瘤坏死因子-α(TNF-α)的影响。方法 将36只Sprague Dawley大鼠随机分为对照组、模型组和姜黄素治疗组,每组12只。模型组和姜黄素治疗组大鼠使用缝扎线结扎冠状动脉前降支制备心肌梗死模型,对照组大鼠冠状动脉前降支下穿线不结扎。各组大鼠均于术前6 d进行灌胃,其中对照组、模型组大鼠于术前6 d给予生理盐水灌胃,每次4 mL,每日1次;姜黄素治疗组大鼠将姜黄素按150 mg·kg-1溶于4 mL体积分数6%的乙醇聚乙二醇400中进行灌胃,每日1次,连续5 d;于术后第6天处死大鼠并取结扎线以下的冠状动脉和心肌组织,反转录-聚合酶链式反应 (RT-PCR)检测各组大鼠心肌组织中TNF-α mRNA表达水平,Western blot检测各组大鼠心肌组织中TNF-α蛋白表达水平,免疫组织化学法检测冠状动脉VEGF表达水平。结果 模型组大鼠心肌组织中TNF-α mRNA和TNF-α蛋白表达水平均显著高于对照组(P<0.05);姜黄素治疗组大鼠心肌组织中TNF-α mRNA和TNF-α蛋白表达水平与模型组比较均显著降低(P<0.05),而与对照组比较差异无统计学意义(P>0.05)。模型组和姜黄素治疗组大鼠结扎线以下冠状动脉组织中VEGF表达水平较对照组显著升高(P<0.05,P<0.01);姜黄素治疗组大鼠结扎线以下冠状动脉组织中VEGF表达水平较模型组明显升高(P<0.01)。结论 姜黄素可有效降低急性心肌梗死大鼠缺血损伤区域心肌组织中TNF-α的表达,并有效提高结扎线以下冠状动脉VEGF的表达,对受损心肌功能的恢复及维持冠状动脉血管内皮功能稳定和血管再生具有积极意义。
Abstract:
Objective To investigate the effect of curcumin on vascular endothelial growth factor(VEGF) of coronary artery and tumor necrosis factor-α(TNF-α)of myocardium tissue in rats with acute myocardial infarction (AMI).Methods Thirty-six Sprague Dawley rats were randomly divided into control group,model group and curcumin treatment group,12 rats in each group.All the rats were given intragastric administration at six days before operation.The rats in control group and model group were given intragastric administration with 4 mL normal saline at six days before operation for five days,once a day.Curcumin was dissolved in 4 mL 6% ethanol polyethylene glycol 400,then the rats in curcumin treatment group were given intragastric administration with the curcumin once a day for five days.All the rats were sacrificed at six days after operation to obtain the coronary artery and myocardium tissue under the ligature.TNF-α mRNA in myocardium tissue was detected by reverse transcription polymerase chain reaction and TNF-α protein in myocardium tissue was detected by Western blot.The change of VEGF in coronary artery was detected by immunohistochemistry method.Results The expression levels of TNF-α mRNA and TNF-α protein in model group were higher than those in control group(P<0.05);while they were lower in curcumin treatment group than those in model group(P<0.05);but there was no significance difference between curcumin treatment group and control group(P>0.05).The VEGF level in coronary artery in curcumin group and model group was higher than that in control group(P<0.05,P<0.01);while the VEGF level in coronary artery in curcumin group was higher than that in model group(P<0.01).Conclusion Curcumin can effectively decrease the expression of TNF-α in ischemic myocardium,and promote the expression of VEGF in coronary artery of rats with acute myocardial infarction,which could promote vessel revascularization and keep the function of endothelia of coronary artery.
