摘要:
目的 探讨PM2.5对大鼠心率变异性(HRV)及心率的影响。方法 将16只Wistar雄性大鼠随机分为空白对照组(n=8)和PM2.5染毒组(n=8),适应性喂养1周后,PM2.5染毒组大鼠给予PM2.5混悬液(10 mg·kg-1·d-1)气管滴注,空白对照组大鼠给予等量生理盐水气管滴注,隔日1次,共3次。末次染毒后24 h麻醉动物,记录大鼠心电图,应用BL-420F生物机能实验系统测定大鼠平均心率及心率变异性(HRV)相关时域和频域指标。结果 PM2.5染毒组大鼠正常窦性RR间期标准差(SDNN)、正常连续窦性RR间期差值均方根(RMSSD)、低频功率(LF)、高频功率(HF)以及低频功率/高频功率(LF/HF)分别为(3.69±0.54)ms、(3.08±0.57)ms、(302.53±36.98)ms2·Hz-1、(42.75±7.10)ms2·Hz-1、7.32±1.07,空白对照组大鼠SDNN、RMSSD、LF、HF及LF/HF分别为(1.68±0.20)ms、(1.69±0.31)ms、(175.69±28.22)ms2·Hz-1、(29.42±3.88)ms2·Hz-1、5.82±1.27,PM2.5染毒组大鼠SDNN、RMSSD、LF、HF及LF/HF均低于空白对照组(P<0.05)。PM2.5染毒组大鼠平均心率为(446.86±25.31)次·min-1,显著快于空白对照组的(359.17±36.27)次·min-1(t=8.361,P<0.05)。结论 PM2.5暴露能够降低大鼠的HRV,加快大鼠的心率。
Abstract:
Objective To explore the effect of PM2.5 on heart rate variability(HRV) and heart rate in rats.Methods Sixteen male Wistar rats were randomly divided into blank control group(n=8)and PM2.5 exposed group(n=8).After adaptive feeding for one week,the rats in PM2.5 exposed group were given PM2.5 suspension(10 mg·kg-1·d-1) by tracheal instillation once every other day for three times;the rats in control group were given the same volume saline by tracheal instillation once every other day for three times.The rats were anesthetized at 24 hours after the last exposure,and the electrocardiograph (ECG) of rats were recorded.The average heart rate and the time domain and frequency domain indexes of HRV in rats were measured by using BL-420F biological function experiment system.Results The standard deviation of normal to normal intervals (SDNN),square root of the mean squared differences of successive normal heart beats (RMSSD),low frequency power (LF),high frequency power (HF) and LF/HF of rats in PM2.5 exposed group was(3.69±0.54)ms,(3.08±0.57)ms,(302.53±36.98)ms2·Hz-1,(42.75±7.10)ms2·Hz-1,7.32±1.07 respectively;the SDNN,RMSSD,LF,HF and LF/HF of rats in blank control group was(1.68±0.20)ms,(1.69±0.31)ms,(175.69±28.22)ms2·Hz-1,(29.42±3.88)ms2·Hz-1,5.82±1.27 respectively;the SDNN,RMSSD,LF,HF and LF/HF of rats in PM2.5 exposed group were significantly less than those in the blank control group(P<0.05).The average heart rate of rats in the PM2.5 exposed group and blank control group was (446.86±25.31),(359.17±36.27)bpm respectively;the average heart rate of rats in the PM2.5 exposed group was significantly higher than that in the control group (t=8.361,P<0.05).Conclusion PM2.5 exposure can reduce the HRV and increase the average heart rate of rat.
