摘要:
目的 探讨泡桐花总黄酮对四氯化碳(CCl4)所致小鼠急性肝损伤的保护作用。方法 50只小鼠随机分为对照组、CCl4模型组、水飞蓟宾对照组、CCl4+黄酮低剂量组和CCl4+黄酮高剂量组,每组10只。对照组和CCl4模型组小鼠给予质量分数1%羧甲基纤维素钠(CMC-Na)10 mL·kg-1混悬液灌胃,水飞蓟宾对照组小鼠给予100 mg·kg-1 水飞蓟宾CMC-Na混悬液灌胃,CCl4+黄酮低剂量组和CCl4+黄酮高剂量组小鼠分别给予泡桐花总黄酮CMC-Na混悬液(100、200 mg·kg-1)灌胃,连续7 d,末次灌胃给药 2 h后,对照组小鼠腹腔注射色拉油0.10 mL·kg-1,其余4组小鼠腹腔注射0.012 mol·L-1CCl4溶液10 mL·kg-1(色拉油配制)造成小鼠急性肝损伤,16 h(禁食不禁水12 h)后摘眼球采血,分离血清,检测血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平及肝脏组织中丙二醛(MDA)和过氧化物歧化酶(SOD)水平。结果 与对照组比较,CCl4模型组小鼠ALT、AST、MDA水平显著升高,SOD水平降低(P<0.01);与CCl4模型组比较,水飞蓟宾对照组、CCl4+黄酮低剂量组和CCl4+黄酮高剂量组小鼠ALT、AST、MDA水平显著降低,SOD水平升高(P<0.05);CCl4+黄酮低剂量组小鼠ALT、AST、MDA、SOD水平与CCl4+黄酮高剂量组比较差异无统计学意义(P>0.05),CCl4+黄酮低、高剂量组与水飞蓟宾对照组比较,差异无统计学意义(P>0.05)。结论 泡桐花总黄酮对CCl4造成的急性肝损伤有明显的保护作用。
Abstract:
Objective To observe the protective effect of flavonoids in Fructus Paulowniae(FFP) on acute liver damage induced by CCl4 in mice.Methods Fifty mice were randomly divided into five groups:control group,CCl4 model group,silibinin control group,CCl4+FFP low dose group and CCl4+FFP high dose group,10 mice in each group.The mice in control group and CCl4 model group were given 1% sodium carboxymethyl cellulose suspension gavage,the mice in silibinin control group were given 100 mg·kg-1 silybin carboxymethylcellulose sodium(CMC-Na) suspension gavage,and the mice in CCl4+FFP low dose group and CCl4+FFP high dose group were given flavonoids in FFP CMC-Na suspension gavage(100,200 mg·kg-1) for 7 consecutive days.After the last intragastric administration of 2 h,the mice in control group were given salad oil 0.10 mL·kg-1 intraperitoneal injection,the mice in other four groups received intraperitoneal injection 10 mL·kg-1 solution of 0.012 mol·L-1 CCl4(salad oil preparation) to induce acute liver injury.Eyeball blood was obtained after 16 h(fasting 12 h),and serum alanine aminotransferase(ALT),aspartate aminotransferase(AST),malondialdehvde(MDA) and superoxide dismutase(SOD) in liver were detected.Results Compared with control group,the level of ALT,AST and MDA in CCl4 model group increased significantly,but level of SOD decreased(P<0.01).Compared with CCl4 model group,the level of ALT,AST and MDA in silibinin control group,CCl4+FFP low dose group and CCl4+FFP high dose group decreased significantly,but the level of SOD increased(P<0.05).There was no significant difference in the level ALT,AST,MDA and SOD between CCl4+FFP low dose group and CCl4+FFP high dose group,and also between CCl4+FFP low dose group,CCl4+FFP high dose group and silibinm control group(P>0.05).Conclusion FFP possess a protective role on CCl4-induced liver damage in mice.
