摘要:
目的 探讨美洲大蠊不同工艺提取物对四氯化碳致急性肝损伤小鼠肝脏的保护作用。方法 72只小鼠随机分为对照组、模型组、水提取组、乙醇提取组、虫粉混悬液组和联苯双酯组,每组12只。采用腹腔注射四氯化碳法建立急性肝损伤小鼠模型。建立模型前,水提取组、乙醇提取组、虫粉混悬液组、联苯双酯组小鼠分别给予美洲大蠊水提取物、乙醇提取物、虫粉混悬液、联苯双酯灌胃,每次200 mg·kg-1,每日1次,连续灌胃10 d;对照组和模型组小鼠给予等量的生理盐水灌胃。建模12 h后称取小鼠体质量,麻醉下摘除眼球采血,采用酶联免疫吸附试验检测血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、总胆红素(TB)水平。颈椎脱臼法处死小鼠,取出肝脏并称质量,计算肝指数;制备肝脏匀浆检测超氧化物歧化酶 (SOD)、丙二醛(MDA)、谷胱甘肽(GSH)活性。结果 模型组小鼠血清ALT、 AST、TB水平显著高于对照组(P<0.01);水提取组、乙醇提取组、虫粉混悬液组和联苯双酯组小鼠血清ALT、 AST、TB水平显著低于模型组(P<0.05);水提取组和虫粉混悬液组小鼠血清ALT、 AST、TB水平显著低于乙醇提取组(P<0.05),水提取组与虫粉混悬液组小鼠血清ALT、 AST、TB水平比较差异无统计学意义(P>0.05),联苯双酯组小鼠血清ALT、 AST、TB水平显著低于水提取组、乙醇提取组和虫粉混悬液组(P<0.05)。模型组小鼠肝脏质量和肝脏指数均显著高于对照组(P<0.05);水提取组、乙醇提取组、虫粉混悬液组、联苯双酯组小鼠肝脏质量和肝脏指数均显著低于模型组(P<0.05);水提取组、乙醇提取组、虫粉混悬液组和联苯双酯组小鼠肝脏质量组间两两比较差异均无统计学意义(P>0.05);水提取组、乙醇提取组、虫粉混悬液组小鼠肝脏指数显著低于联苯双酯组(P<0.05),水提取组和虫粉混悬液组小鼠肝脏指数显著低于乙醇提取组(P<0.05),水提取组与虫粉混悬液组小鼠肝脏指数比较差异无统计学意义(P>0.05)。模型组小鼠肝组织中MDA水平显著高于对照组(P<0.01),SOD和GSH水平显著低于对照组(P<0.01)。水提取组、乙醇提取组、虫粉混悬液组和联苯双酯组小鼠肝组织中MDA水平显著低于模型组(P<0.05),SOD和GSH水平显著高于模型组(P<0.05)。水提取组、乙醇提取组、虫粉混悬液组小鼠肝组织中MDA水平显著高于联苯双酯组(P<0.05),SOD水平显著低于联苯双酯组(P<0.05);水提取组和虫粉混悬液组小鼠肝组织中MDA水平显著低于乙醇提取组(P<0.05),SOD和GSH水平显著高于乙醇提取组(P<0.05)。水提取组与虫粉混悬液组小鼠肝组织中MDA、SOD及GSH水平比较差异均无统计学意义(P>0.05)。乙醇提取组小鼠肝组织中GSH水平显著低于联苯双酯组(P<0.05),水提取组、虫粉混悬液组和联苯双酯组小鼠肝组织中GSH水平比较差异均无统计学意义(P>0.05)。结论 美洲大蠊提取物对小鼠急性肝损伤有保护作用,其中美洲大蠊虫粉及其水提取物效果最好,乙醇提取物效果次之。
Abstract:
Objective To study the protective effect of Periplaneta americana extracts extracted by different technique on acute hepatic injury induced by carbon tetrachloride(CCl4)in mice.Methods A total of 72 mice were randomly divided into control group,model group,water extraction group,ethanol extraction group,insect powder suspension group and bifendate groups,12 mice in each group.The mouse models of acute hepatic injury were established by intraperitoneal injection of carbon tetrachloride.Before the establishment of the models,the mice in water extraction group,ethanol extraction group,insect powder suspension group and bifendate groups were treated with Periplaneta americana water extract,ethanol extract,insect powder suspension and bifendate 200 mg·kg-1 by intragastric administration,once a day for ten days;the mice in control group and model group were treated with equal volume of normal saline by intragastric administration.The body weight of mice was weighed 12 hours after modeling.The blood was collected by enucleating the eyeball.The levels of serum alanine transarninase(ALT),aspartate aminotransferase(AST)and total bilirubin(TB)were detected by enzyme-linked immunosorbent test.The livers were enucleated and weighed after the mice were killed by cervical dislocation,then the liver index was calculated.The levels of superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione(GSH)in hepatic tissue homogenate were detected.Results The levels of serum ALT,AST and TB in model group were significantly higher than those in control group(P<0.01).The levels of serum ALT,AST and TB in water extraction group,ethanol extraction group,insect powder suspension group and bifendate group were significantly lower than those in model group(P<0.05).The levels of serum ALT,AST and TB in water extraction group and insect powder suspension group were significantly lower than those in ethanol extraction group(P<0.05);but there was no significant difference in the levels of serum ALT,AST and TB between water extraction group and insect powder suspension group(P>0.05).The levels of serum ALT,AST and TB in bifendate group were significantly lower than those in water extraction group,ethanol extraction group and insect powder suspension group(P<0.05).The liver mass and liver index in model group were significantly higher than those in control group(P<0.05).The liver mass and liver index in water extraction group,ethanol extraction group,insect powder suspension group and bifendate group were significantly lower than those in model group(P<0.05).There was no significant difference in the liver mass between the any two groups of water extraction group,ethanol extraction group,insect powder suspension group and bifendate group(P>0.05).The liver index of mice in water extraction group,ethanol extraction group and insect powder suspension group was significantly lower than that in bifendate group(P<0.05).The liver index of mice in water extraction group and insect powder suspension group was significantly lower than that in ethanol extraction group(P<0.05).But there was no significant difference in the liver index of mice between water extraction group and insect powder suspension group(P>0.05).The MDA level in hepatic tissue of mice in model group was significantly higher than that in control group(P<0.01),but the levels of SOD and GSH in model group was significantly lower than that in control group(P<0.01).The MDA level in hepatic tissue of mice in water extraction group,ethanol extraction group,insect powder suspension group and bifendate group was significantly lower than that in model group(P<0.05);but the levels of SOD and GSH in hepatic tissue of mice in water extraction group,ethanol extraction group,insect powder suspension group and bifendate group were significantly higher than those in model group(P<0.05).The MDA level in hepatic tissue of mice in water extraction group,ethanol extraction group and insect powder suspension group was significantly higher than that in bifendate group(P<0.05);but the SOD level in hepatic tissue of mice in water extraction group,ethanol extraction group and insect powder suspension group was significantly lower than that in bifendate group(P<0.05).The MDA level in hepatic tissue of mice in water extraction group and insect powder suspension group was significantly lower than that in ethanol extraction group(P<0.05),but the MDA level in hepatic tissue of mice in water extraction group and insect powder suspension group was significantly higher than that in ethanol extraction group(P<0.05).There was no significant difference in the levels of MDA,SOD and GSH in hepatic tissue of mice between water extraction group and insect powder suspension group(P>0.05).The GSH level in hepatic tissue of mice in ethanol extraction group was significantly lower than that in bifendate group(P<0.05).There was no significant difference in the GSH level in hepatic tissue of mice between water extraction group,insect powder suspension group and bifendate group(P>0.05).Conclusion Periplaneta americana extracts has protective effect on acute hepatic injury of mice,and the effects of Periplaneta americana powder and its water extract are better than its ethanol extract.
