摘要:
目的 探讨益气清热活血方的浸渍剂与巴布膏剂对创伤性肌肉损伤兔肌肉组织中水通道蛋白-1(AQP-1)及血清中白细胞介素-8(IL-8)表达水平的影响。方法 将40只新西兰雄兔在适宜环境下饲养1周后随机分为空白组、模型组、浸渍组和巴布膏组,每组10只。空白组兔不给予任何干预措施;模型组、浸渍组和巴布膏组兔制备创伤性肌肉损伤模型。造模前对各组兔进行脱毛,造模成功后40 min,模型组兔用不含药物的巴布膏贴剂贴于患处10 h,每日1次;浸渍组兔用棉签蘸取适量清热益气活血浸渍液涂擦于患处,每日3次;巴布膏组兔用清热益气活血巴布膏贴剂贴于患处10 h,每日1次。分别于脱毛后、造模后及治疗24、72 h时对各组兔进行损伤症状评分。治疗24、72 h时,采用酶联免疫吸附试验法检测各组兔血清中IL-8水平,采用荧光定量聚合酶链式反应法检测各组兔肌肉组织中AQP-1 mRNA的相对表达量。结果 脱毛后4组兔损伤症状评分比较差异均无统计学意义(P>0.05)。造模后和治疗24、72 h时,模型组、浸渍组和巴布膏组兔损伤症状评分显著高于空白组(P<0.05);造模后、治疗24 h时,模型组、浸渍组和巴布膏组兔损伤症状评分比较差异均无统计学意义(P>0.05);治疗72 h时,浸渍组和巴布膏组兔损伤症状评分显著低于模型组(P<0.05),浸渍组与巴布膏组兔损伤症状评分比较差异无统计学意义(P>0.05)。空白组兔不同时间点损伤症状评分比较差异无统计学意义(P>0.05)。模型组、浸渍组、巴布膏组兔造模后及治疗24、72 h 时的损伤症状评分均显著高于脱毛后(P<0.05)。模型组兔治疗24、72 h时的损伤症状评分与造模后比较差异无统计学意义(P>0.05);模型组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。浸渍组兔治疗24 h 时的损伤症状评分与造模后比较差异无统计学意义(P>0.05);浸渍组兔治疗72 h时的损伤症状评分显著低于造模后(P<0.05);浸渍组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。巴布膏组兔治疗24、72 h时的损伤症状评分显著低于造模后(P<0.05);巴布膏组兔治疗72 h时的损伤症状评分显著低于治疗24 h时(P<0.05)。 治疗24、72 h时,模型组、浸渍组、巴布膏组兔血清中IL-8水平均显著高于空白组(P<0.05)。治疗24 h时,浸渍组与模型组兔血清中IL-8水平比较差异无统计学意义(P>0.05),巴布膏组兔血清中IL-8水平显著低于模型组(P<0.05),巴布膏组与浸渍组兔血清中IL-8水平比较差异无统计学意义(P>0.05)。治疗72 h时,浸渍组、巴布膏组兔血清中IL-8水平显著低于模型组(P<0.05),巴布膏组与浸渍组兔血清中IL-8水平比较差异无统计学意义(P>0.05)。治疗24、72 h时,模型组、浸渍组、巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量显著高于空白组(P<0.05)。治疗24 h时,模型组、浸渍组、巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量比较差异均无统计学意义(P>0.05)。治疗72 h时,浸渍组和巴布膏组兔肌肉组织中AQP-1 mRNA相对表达量显著低于模型组(P<0.05),巴布膏组与浸渍组兔肌肉组织中AQP-1 mRNA相对表达量比较差异无统计学意义(P>0.05)。空白组、模型组兔治疗24 h时与治疗72 h时血清中IL-8水平和肌肉组织中AQP-1 mRNA相对表达量比较差异均无统计学意义(P>0.05)。浸渍组、巴布膏组兔治疗72 h时血清中IL-8水平和肌肉组织中AQP-1 mRNA相对表达量显著低于治疗24 h时(P<0.05)。 结论 创伤性肌肉损伤兔在自然条件下72 h后可缓慢修复,清热益气活血方的浸渍剂与巴布膏剂均可有效加快其修复,该作用可能与抑制AQP-1及IL-8的表达有关。
Abstract:
Objective To investigate the effect of the immersion liquid and the Babu ointment of Yiqi Qingre Huoxue decoction on the expression of aquaporin-1(AQP-1) in muscle tissues and the interleukin-8(IL-8)level in serum of rabbits with traumatic muscle injury.Methods Forty New Zealand male rabbits were randomly divided into blank group,model group,immersion liquid group and Babu ointment group after being raised for one week in suitable environment,with 10 rabbits in each group.The rabbits in the blank group were not given any intervention;the rabbits in the model group,immersion liquid group and Babu ointment group were made the traumatic muscle injury model.The rabbits in each group were depilated before modeling;at 40 minutes after the successful modeling,the Babu ointment plaster without drugs were applied to the affected area of the rabbits in the model group for 10 hours,once a day;the cotton swabs were applied to dip a proper amount of Yiqi Qingre Huoxue solution to the affected area of the rabbits in the immersion liquid group,three times a day;the Babu ointment patch of Yiqi Qingre Huoxue decoction was applied to the affected area of the rabbits in the Babu ointment group for 10 hours,once a day.The injury symptom scores of rabbits in each group were evaluated after depilation,after modeling,and at 24 and 72 hours of treatment.At 24 and 72 hours of treatment,the level of IL-8 in serum of rabbits in each group was detected by enzyme-linked immunosorbent assay,and the relative expression of AQP-1 mRNA in muscle tissue of rabbits in each group was detected by fluorescence quantitative polymerase chain reaction method.Results There was no significant difference in the injury symptom score among the four groups after depilation (P>0.05).After modeling and at 24,72 hours of treatment,the injury symptom score of rabbits in the model group,immersion liquid group and Babu ointment group were significantly higher than those in the blank group(P<0.05).After modeling and at 24 hours of treatment,there was no significant difference in the injury symptom score of rabbits among the model group,immersion liquid group and Babu ointment group(P>0.05).At 72 hours of treatment,the injury symptom scores of rabbits in the immersion liquid group and Babu ointment group were significantly lower than those in the model group(P<0.05);there was no significant difference in the injury symptom score of rabbits between the immersion liquid group and Babu ointment group(P>0.05).There was no significant difference in injury symptom score of rabbits at different time points in the blank group(P>0.05).The injury symptom scores of rabbits after modeling and at 24 and 72 hours of treatment were significantly higher than those of depilation in the model group,immersion liquid group and Babu ointment group(P<0.05).There was no significant difference in the injury symptom score of rabbits at 24,72 hours of treatment and after modeling in the model group(P>0.05);the injury symptom score of rabbits at 72 hours of treatment was significantly lower than that at 24 hours of treatment in the model group(P<0.05).There was no significant difference in the injury symptom scores of rabbits at 24 hours of treatment and after modeling in the immersion liquid group(P>0.05);the injury symptom score of rabbits at 72 hours of treatment was significantly lower than that after modeling in the immersion liquid group(P<0.05);the injury symptom score of rabbits at 72 hours of treatment was significantly lower than that at 24 hours of treatment in the immersion liquid group(P<0.05).The injury symptom scores of rabbits at 24,72 hours of treatment were significantly lower than those after modeling in the Babu ointment group(P<0.05);the injury symptom score of rabbits at 72 hours of treatment was significantly lower than that at 24 hours of treatment in the Babu ointment group(P<0.05).At 24,72 hours of treatment,the level of IL-8 in serum of rabbits in model group,immersion liquid group and Babu ointment group was significantly higher than that in the blank group(P<0.05).At 24 hours of treatment,there was no significant difference in serum IL-8 level between the immersion liquid group and the model group(P>0.05);the serum IL-8 level of rabbits in Babu ointment group was significantly lower than that in the model group(P<0.05);there was no significant difference in serum IL-8 level between the Babu ointment group and the immersion liquid group(P>0.05).At 72 hours of treatment,the level of IL-8 in serum of rabbits in the immersion liquid group and Babu ointment group was significantly lower than that in the model group(P<0.05);there was no significant difference in serum IL-8 level between the Babu ointment group and the immersion liquid group(P>0.05).At 24,72 hours of treatment,the relative expression of AQP-1 mRNA in muscle tissue of rabbits in the model group,immersion liquid group and Babu ointment group was significantly higher than that in the blank group(P<0.05).At 24 hours of treatment,there was no significant difference in the relative expression of AQP-1 mRNA in muscle tissues among the model group,immersion liquid group and Babu ointment group(P>0.05).At 72 hours of treatment,the relative expression of AQP-1 mRNA in muscle tissue of rabbits in the immersion liquid group and Babu ointment group was significantly lower than that in the model group(P<0.05);there was no significant difference in the relative expression of AQP-1 mRNA between the Babu ointment group and the immersion liquid group(P>0.05).There was no significant difference in serum IL-8 level and the relative expression of AQP-1 mRNA in muscle tissue of rabbits at 24 hours of treatment and 72 hours of treatment in the blank group and the model group(P>0.05).The level of IL-8 in serum and the relative expression of AQP-1 mRNA in muscle tissue of rabbits at 72 hours of treatment were significantly lower than those at 24 hours of treatment in the immersion liquid group and Babu ointment group(P<0.05).Conclusion The traumatic muscle injury of rabbit can be slowly repaired after 72 hours under natural conditions.Both the immersion liquid and the Babu ointment of Yiqi Qingre Huoxue decoction can effectively speed up the repair of the injured muscle,which may be related to the inhibition of the expression of AQP-1 and IL-8.
