摘要:
目的 探讨miRNA-505-3p在原发性高血压(EH)发生机制中的作用。方法 选择2014年5~8月于陆军军医大学附属第二医院收治的EH患者和非EH患者各60例,采集患者静脉血,分离血浆,采用实时荧光定量聚合酶链反应(PCR)检测血浆中miRNA-505-3p相对表达量,采用酶联免疫吸附试验检测血浆中胰岛素样生长因子-1(IGF-1)水平。培养人主动脉平滑肌细胞(HASMC),当细胞融合度达到50%~70%时随机分为mimic组(转染miRNA-505-3p mimic序列)、mimic对照组(转染mimic对照序列)和空白对照组(不进行转染);转染后继续培养72 h,然后收集细胞及细胞培养液。采用实时荧光定量PCR检测3组HASMC中miRNA-505-3p及IGF-1 mRNA表达,细胞计数试剂盒-8检测3组HASMC的活性,Western blot法检测3组HASMC中IGF-1蛋白表达。结果 EH患者与非EH患者血浆中miRNA-505-3p相对表达量分别为0.156 5±0.025 9、0.007 9±0.001 4,EH患者血浆中miRNA-505-3p相对表达量显著高于非EH患者(t=6.880,P<0.01)。EH患者和非EH患者血浆miRNA-505-3p的受试者工作特征曲线下面积为0.988 1,95%可信区间为0.972 4~0.999 0(P<0.01)。EH患者和非EH患者血浆中IGF-1水平分别为(1 338.03±96.92)、(620.90±23.19)ng·L-1,EH患者血浆中IGF-1水平显著高于非EH患者(t=7.101,P<0.01)。mimic组、mimic对照组和空白对照组HASMC活性分别为0.679 7±0.032 8、0.471 0±0.024 5、0.525 2±0.035 3;mimic组HASMC活性显著高于mimic对照组和空白对照组(t=6.203、4.610,P<0.05),mimic对照组与空白对照组HASMC活性比较差异无统计学意义(t=1.432,P>0.05)。mimic组、mimic对照组和空白对照组HASMC中miRNA-505-3p相对表达量分别为 0.693 3±0.022 9、0.020 6±0.000 8、0.023 8±0.001 1,mimic组HASMC中miRNA-505-3p相对表达量显著高于mimic对照组和空白对照组(t=7.991、5.840,P<0.01),mimic对照组与空白对照组HASMC中miRNA-505-3p相对表达量比较差异无统计学意义(t=0.323,P>0.05)。mimic组、mimic对照组和空白对照组HASMC中IGF-1 mRNA相对表达量分别为0.238 6±0.013 3、0.082 4±0.004 0、0.079 7±0.004 7,mimic组HASMC中IGF-1 mRNA相对表达量显著高于mimic对照组和空白对照组(t=16.903、18.240,P<0.01),mimic对照组与空白对照组HASMC中IGF-1 mRNA相对表达量比较差异无统计学意义(t=0.295,P>0.05)。mimic组、mimic对照组和空白对照组HASMC中IGF-1蛋白相对表达量分别为0.602 0±0.017 3、0.040 3±0.006 2、0.051 6±0.001 0;mimic组HASMC中IGF-1蛋白相对表达量显著高于mimic对照组和空白对照组(t=29.026、31.557,P<0.01),mimic对照组与空白对照组HASMC中IGF-1蛋白相对表达量比较差异无统计学意义(t=0.516,P>0.05)。结论 EH患者血浆miRNA-505-3p表达上调,miRNA-505-3p可能通过调控IGF-1表达而参与EH的发病,并可作为EH的生物学标志物。
Abstract:
Objective To investigate the role of miRNA-505-3p in the pathogenesis of essential hypertension (EH).Methods Sixty EH patients and sixty non-EH patients were selected from May to August 2014 in the Second Affiliated Hospital of Army Medical University.The venous blood samples were collected,and the plasma was separated.The expression of miRNA-505-3p in plasma of EH patients and non-EH patients was detected by real-time fluorescence quantitative polymerase chain reaction (PCR);the level of insulin-like growth factor-1 (IGF-1) in plasma was detected by enzyme linked immunosorbent assay.The human aortic smooth muscle cells (HASMC) were cultured,and the cells were randomly divided into mimic group (transfected with miRNA-505-3p mimic sequence),mimic-control group (transfected with mimic control sequence) and blank control group (not transfected) when the cell fusion reached 50%-70%.The cells were cultured for 72 hours after transfection,then the cells and cell culture medium were collected.The expression of miRNA-505-3p and IGF-1 mRNA in HASMC of the three groups was detected by real-time fluorescence quantitative PCR,the activity of HASMC was detected by cell counting kit-8,and the expression of IGF-1 protein in HASMC was detected by Western blot.Results The relative expression of plasma miRNA-505-3p in EH patients and non-EH patients was (0.156 5±0.025 9) and (0.007 9±0.001 4),respectively.The relative expression of plasma miRNA-505-3p in EH patients was significantly higher than that in non-EH patients (t=6.880,P<0.01).The area under the receiver operating characteristic curve of plasma miRNA-505-3p in EH patients and non-EH patients was 0.988 1,and the 95% confidence interval was 0.972 4-0.999 0 (P<0.01).The plasma IGF-1 level in EH patients and non-EH patients was (1 338.03±96.92) and (620.90±23.19) ng·L-1,respectively.The plasma IGF-1 level in EH patients were significantly higher than that in non-EH patients (t=7.101,P<0.01).The activity of HASMC in the mimic group,mimic-control group and blank control group was 0.679 7±0.032 8,0.471 0±0.024 5 and 0.525 2±0.035 3,respectively.The activity of HASMC in the mimic group was significantly higher than that in the mimic-control group and blank control group (t=6.203,4.610;P<0.05),but there was no significant difference in the activity of HASMC between the mimic-control group and blank control group (t=1.432,P>0.05).The relative expression of miRNA-505-3p in HASMC of the mimic group,mimic-control group and blank control group was 0.693 3±0.022 9,0.020 6±0.000 8 and 0.023 8±0.001 1,respectively.The relative expression of miRNA-505-3p in HASMC of the mimic group was significantly higher than that of the mimic-control group and blank control group (t=7.991,5.840;P<0.01).There was no significant difference in the relative expression of miRNA-505-3p in HASMC between the mimic-control group and blank control group (t=0.323,P>0.05).The relative expression of IGF-1 mRNA in HASMC of the mimic group,mimic-control group and blank control group was 0.238 6±0.013 3,0.082 4±0.004 0 and 0.079 7±0.004 7,respectively.The relative expression of IGF-1 mRNA in HASMC of the mimic group was significantly higher than that of mimic-control group and blank control group (t=16.903,18.240;P<0.01).There was no significant difference in the relative expression of IGF-1 mRNA between the mimic-control group and blank control group (t=0.295,P>0.05).The relative expression of IGF-1 protein in HASMC of the mimic group,mimic-control group and blank control group was 0.602 0±0.017 3,0.040 3±0.006 2 and 0.051 6±0.001 0,respectively.The relative expression of IGF-1 protein in HASMC of the mimic group was significantly higher than that of mimic-control group and blank control group (t=29.026,31.557;P<0.01).There was no significant difference in the relative expression of GF-1 protein between the mimic-control group and blank control group (t=0.516,P>0.05).Conclusion The expression of miRNAs-505-3p in plasma of EH patients is up-regulated.MiRNAs-505-3p may participate in the pathogenesis of EH by regulating the expression of IGF-1,and may be used as a biomarker of EH.
