摘要:
目的 探讨不同浓度替比夫定对大鼠肾小球系膜细胞(RMCs)增殖的影响。方法 将RMCs分为空白对照组和10、20、40、60、100、150 mmol·L-1替比夫定组,分别作用24、48、72 h,通过细胞形态学和4-甲基偶氮唑蓝法检测替比夫定对RMCs增殖的影响。结果 空白对照组细胞胞质透亮,折光性强,细胞呈梭形、不规则星形或多角形,细胞密集时可呈网状排列。10、40和150 mmol·L-1替比夫定组细胞形态随药物浓度增加及作用时间的延长出现皱缩,细胞核缩成点状,甚至出现胞质空泡。10、20、40、60、100和150 mmol·L-1替比夫定组作用RMCs 24、48、72 h后,RMCs增殖抑制率随药物浓度的增加和作用时间的延长而增高,且呈剂量和时间依赖性(P<0.01)。结论 替比夫定对RMCs有显著的抑制作用,并且具有时间和剂量依赖性。
Abstract:
Objective To study the effect of telbivudine on the proliferation of rat mesangial cells(RMCs) cultured in vitro.Methods RMCs were divided into blank control group and telbivudine group.In telbivudine group,RMCs were treated with different concentrations of telbivudine (10,20,40,60,100,150 mmol·L-1).After treatment for 24,48 and 72 hours,the cell proliferation was observed by morphological and evaluated by methylthiazolyl tetrazolium assay.Results In blank control group,cytoplasm of RMCs was lucency,refractivity was better,the shape of the cells appeared fusiformis,irregular star-shaped and polygon.The cells showed reticulate arrangement when the cells was dense.The RMCs in 10,40 and 150 mmol·L-1 telbivudine groups reacted with the increasing of telbivudine for concentration and time,cell morphology atrophy appeared,the nucleus shrink to a dot and apart of cells appeared some vacuoles.The RMCs in 10,20,40,60,100 and 150 mmol·L-1 telbivudine groups were treated by telbivudine for 24,48 and 72 hours,proliferation inhibition ratio increased with the increase of concentration and time,and showed a dose and time dependence(P<0.01).Conclusion telbivudine has significant inhibitory effect on RMCs with dose and time dependence.
