摘要:
目的 探讨七氟烷后处理对大鼠心肌缺血再灌注损伤的影响。方法 采用随机数字表法将30只健康成年雄性Sprague Dawley大鼠分为对照组、缺血再灌注组和七氟烷组,每组10只。缺血再灌注组和七氟烷组大鼠制备缺血再灌注损伤模型。缺血再灌注组大鼠冠状动脉结扎30 min后,拔出栓线再灌注120 min;七氟烷组大鼠于再灌注前1 min吸入体积分数2%七氟烷15 min,其余同缺血再灌注组;对照组大鼠步骤同缺血再灌注组,但不结扎冠状动脉。 麻醉大鼠后,应用多功能生物信号采集系统监测并记录大鼠开胸前(T0)、缺血10 min(T1)、缺血30 min(T2)、再灌注30 min(T3)、再灌注60 min(T4)、再灌注120 min(T5)时的心率(HR)和平均动脉压(MAP)。采用酶联免疫吸附试验检测3组大鼠缺血再灌注120 min后血清中心肌肌钙蛋白I(cTnI)、肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)水平。结果 T0、T1、T2时3组大鼠的HR和MAP比较差异无统计学意义(P>0.05);T3、T4、T5时缺血再灌注组和七氟烷组大鼠的HR和MAP显著低于对照组 (P<0.05);T3、T4、T5时七氟烷组大鼠的MAP显著高于缺血再灌注组(P<0.05);T3、T4、T5时缺血再灌注组与七氟烷组大鼠的HR比较差异无统计学意义(P>0.05)。对照组大鼠T1、T2、T4时的HR显著低于T0时,T3、T5时的HR显著高于T0时,T2、T3、T4、T5时的MAP显著低于T0时,T2时的HR显著低于T1时,T3、T4、T5时的HR显著高于T1时,T2、T3、T4、T5时的MAP显著低于T1时,T3、T4、T5时的HR显著高于T2时,T3、T5时的MAP显著低于T2时,T4时的MAP显著高于T2时(P<0.05)。对照组大鼠 T3、T4、T5时的MAP比较差异无统计学意义(P>0.05)。缺血再灌注组大鼠中,T2、 T3、T4、T5时的HR和MAP显著低于T0、T1时,T3、T4、T5时的HR和MAP显著低于T2时,T4、T5时的HR显著低于T3时,T5时的MAP显著低于T3时,T5时的HR和MAP显著低于T4时 (P<0.05)。七氟烷组大鼠中,T1、T2、 T3、T4、T5时的HR、MAP显著低于T0时,T2、 T3、T4、T5时的HR、MAP显著低于T1时,T3、T4、T5时的HR、MAP显著低于T2时 (P<0.05);七氟烷组大鼠T3、T4、T5时的HR、MAP比较差异无统计学意义(P>0.05)。缺血再灌注组和七氟烷组大鼠血清中cTnI、LDH、CK-MB水平均显著高于对照组 (P<0.05);七氟烷组大鼠血清中cTnI、LDH、CK-MB水平均显著低于缺血再灌注组 (P<0.05)。结论 七氟烷后处理可改善缺血再灌注损伤大鼠心肌的血流供应,降低血清cTnI、LDH和CK-MB水平,对缺血再灌损伤的心肌有一定的保护作用。
Abstract:
Objective To explore the effect of sevoflurane post-treatment on myocardial ischemia-reperfusion injury of rats.Methods Thirty healthy adult male Sprague Dawley rats were divided into control group,ischemia-reperfusion group and sevoflurane group by random number table method,with 10 rats in each group.Rats in the ischemia-reperfusion group and sevoflurane group were prepared for ischemia-reperfusion injury models.After 30 minutes of coronary artery ligation of rats in the ischemia-reperfusion group,the thrombus was pulled out and reperfused for 120 minutesthe rats in the sevoflurane group were inhaled with a volume fraction of 2% sevoflurane at 1 minute before reperfusion for 15 minutes,and the rest of the steps were the same as ischemia-reperfusion groupthe rats in the control group had the same procedure as the ischemia-reperfusion group,but the coronary arteries were not ligated.After the rats were anesthetized,the heart rate (HR) and mean arterial pressure (MAP) of rats were monitored and recorded by the multifunctional biosignal acquisition system before chest opening (T0) and at ischemia 10 minutes (T1),ischemia 30 minutes (T2),reperfusion 30 minutes (T3),reperfusion 60 minutes (T4) and reperfusion 120 minutes (T5).The serum cardiac troponin I(cTnI),creatine kinase isoenzyme (CK-MB) and lactate dehydrogenase (LDH) of rats in the three groups were detected by the enzyme-linked immunosorbent assay after ischemia reperfusion for 120 minutes.Results There was no significant difference in the HR and MAP among the three groups of rats at T0,T1 and T2 (P>0.05)the HR and MAP of rats in the ischemia-reperfusion group and sevoflurane group at T3,T4,T5 were significantly lower than those in the control group (P<0.05)the MAP of rats in the sevoflurane group at T3,T4 and T5 were significant higher than that in the ischemia-reperfusion group (P<0.05)there was no significant difference in the HR between the the ischemia-reperfusion group and sevoflurane group at T3,T4 and T5 (P>0.05).In the control group,the HR of rats at T1,T2,T4 was significantly lower than that at T0,the HR of rats at T3,T5 was significantly higher than that at T0,the MAP of rats at T2,T3,T4,T5 was significantly lower than at that at T0,the HR of rats at T2 was significantly lower than that at T1,and the HR of rats at T3,T4,T5 was significantly higher than that at T1,the MAP of rats at T2,T3,T4,T5 was significantly lower than that at T1,the HR of rats at T3,T4,T5 was significantly higher than that at at T2,the MAP of rats at T3 and T5 was significantly lower than that at T2,the MAP of rats at T4 was significantly higher than that at T2 (P<0.05).There was no significant difference in the MAP of rats in the control group among T3,T4 and T5(P>0.05).In the ischemia-reperfusion group,the HR and MAP of rats at T2,T3,T4,T5 were significantly lower than those at T0 and T1,the HR and MAP of rats at T3,T4,T5 were significantly lower than those at T2,the HR of rats at T4 and T5 was significantly lower than that at T3,the MAP of rats at T5 was significantly lower than those at T3,the HR and MAP of rats at T5 were significantly lower than those at T4 (P<0.05).In the sevoflurane group,the HR and MAP of rats at T1,T2,T3,T4,T5were significantly lower than those at T0,the HR and MAP of rats at T2,T3,T4,T5 were significantly lower than those at T1,the HR and MAP of rats at T3,T4,T5 were significantly lower than those at T2 (P<0.05)there was no significant difference in the HR and MAP of rats in the sevoflurane group among T3,T4,T5 (P>0.05).the levels of cTnI,LDH and CK-MB in the serum of rats in the ischemia-reperfusion group and sevoflurane group were significantly higher than those in the control group (P<0.05)the levels of cTnI,LDH and CK-MB in the serum of rats in the sevoflurane group were significantly lower than those in the ischemia-reperfusion group (P<0.05).Conclusion Sevoflurane post-treatment can improve the blood flow supply of myocardium of rats with ischemia-reperfusion injury,reduce cTnI,LDH and CK-MB levels in serum,and have a certain protective effect on myocardial ischemia-reperfusion injury.
