摘要:
目的 探讨小分子RNA干扰磷脂酰肌醇3-激酶催化亚基α(PIK3CA)基因对人骨肉瘤Saos-2细胞增殖和侵袭能力的影响。方法 培养人骨肉瘤Saos-2细胞,将细胞随机分为siRNA-PIK3CA组、siRNA对照组和空白对照组,siRNA-PIK3CA组细胞转染siRNA-PIK3CA序列,siRNA对照组细胞转染siRNA对照序列,空白对照组细胞正常培养,不作任何干预。实时荧光定量聚合酶链反应检测各组细胞中PIK3CA mRNA表达,四甲基偶氮唑盐(MTT)比色法检测各组细胞增殖情况,流式细胞术检测各组细胞凋亡情况,Transwell法检测各组细胞迁移和侵袭能力,Western blot法检测各组细胞中PIK3CA、磷酸肌醇3激酶(PI3K)、磷酸化-PI3K (p-PI3K)、蛋白激酶B(Akt)、磷酸化-Akt(p-Akt)、哺乳动物雷帕霉素靶蛋白(mTOR)和磷酸化- mTOR(p-mTOR)蛋白表达。结果 siRNA-PIK3CA组细胞中PIK3CA mRNA和蛋白相对表达量均低于siRNA对照组和空白对照组(P<0.05);siRNA-PIK3CA组细胞转染后24、48、72、96 h 增殖能力均低于siRNA对照组和空白对照组(P<0.05);siRNA-PIK3CA组细胞凋亡率高于siRNA对照组和空白对照组(P<0.05);siRNA-PIK3CA组迁移细胞数和侵袭细胞数均少于siRNA对照组和空白对照组(P<0.05);siRNA-PIK3CA组细胞中PI3K、Akt、mTOR蛋白相对表达量均高于siRNA对照组和空白对照组(P<0.05),而p-PI3K、p-Akt、p-mTOR蛋白相对表达量均低于siRNA对照组和空白对照组(P<0.05)。siRNA对照组与空白对照组细胞中PIK3CA mRNA和蛋白相对表达量、细胞增殖能力、迁移细胞数和侵袭细胞数及PI3K、Akt、mTORp-PI3K、p-Akt、p-mTOR蛋白相对表达量比较差异均无统计学意义(P>0.05)。结论 下调PIK3CA基因可减少人骨肉瘤细胞增殖,抑制细胞迁移及侵袭能力,增加细胞凋亡,其机制可能与抑制PI3K/Akt/mTOR信号通路有关。
Abstract:
Objective To investigate the effects of small RNA interference phosphatidylinositol 3-kinase catalytic subunit alpha (PIK3CA) on the proliferation and invasion of human osteosarcoma cell line Saos-2.Methods The human osteosarcoma cell line Saos-2 were cultured.All cells were randomly divided into siRNA-PIK3CA group,siRNA control group and blank control group.Cells in the siRNA-PIK3CA group were transfected with siRNA-PIK3CA sequence,in the siRNA control group were transfected with siRNA control sequences,and in the blank control group were cultured normally without any intervention.The expression of PIK3CA mRNA in each transfection group was detected by real-time fluorescent quantitative polymerase chain reaction.The cell proliferation in each group was detected by methyl thiazolyl tetrazolium (MTT) assay.The cell apoptosis in each group was detected by flow cytometry.The cell migration and invasion abilities in each group were detected by Transwell method.The expressions of PIK3CA,phosphoinositide 3-kinase (PI3K),phosphorylation-PI3K (p-PI3K),protein kinase B (Akt),phosphorylation-Akt (p-Akt),mammalian target of rapamycin protein (mTOR) and phosphorylation-mTOR (p-mTOR) in each group were detected by Western blot.Results The relative expression levels of PIK3CA mRNA and protein in siRNA-PIK3CA group were lower than those in the siRNA control group and blank control group (P<0.05).The absorbance values at 24,48,72,96 hours after transfection in the siRNA-PIK3CA group were lower than those in the siRNA control group and blank control group (P<0.05).The apoptosis rate in the siRNA-PIK3CA group was higher than that in the siRNA control group and blank control group (P<0.05).The number of migrating cells and invasive cells in siRNA-PIK3CA group were significantly lower than those in the siRNA control group and blank control group (P<0.05).The relative expression levels of PI3K,Akt and mTOR protein in the siRNA-PIK3CA group were higher than those in the siRNA control group and blank control group(P<0.05),while the relative expression levels of p-PI3K,p-Akt and p-mTOR protein were lower than those in the siRNA control group and blank control group (P<0.05).The differences of relative expression levels of PIK3CA mRNA and protein,cell proliferation,the number of migration cells and the number of invasive cells,the relative expression levels of PI3K,Akt,mTORp-PI3K,p-Akt,p-mTOR proteins between siRNA control group and blank control group were not statistically significant (P>0.05).Conclusion Down-regulation of PIK3CA gene can reduce human osteosarcoma cell proliferation,inhibit cell migration and invasion and increase cell apoptosis.The mechanism may be related to the inhibition of PI3K/Akt/mTOR signaling pathway.
