摘要:
目的 观察外源性胆红素对缺血缺氧性脑损伤大鼠认知功能及海马组织中蛋白酪氨酸激酶2(JAK2)/信号转导子与激活子3(STAT3)/B细胞淋巴瘤基因-2(Bcl-2)通路的影响。方法 将72只无特定病原体级7日龄雄性Wistar大鼠随机分为假手术组、模型组、地塞米松组及外源性胆红素低、中、高剂量组,每组12只。各组大鼠腹腔注射水合氯醛麻醉,分离左侧颈总动脉,用丝线结扎颈外动脉(假手术组大鼠只穿线不结扎),模型制备4 h后,地塞米松组大鼠腹腔内注射10 mg·kg-1地塞米松,外源性胆红素低、中、高剂量组大鼠分别腹腔内注射5、10、15 mg·kg-1外源性胆红素,假手术组、模型组大鼠腹腔内注射等量的生理盐水,连续2周。采用水迷宫实验检测各组大鼠的认知功能,末端脱氧核苷酸转移酶介导的 dUTP 缺口末端标记法检测各组大鼠海马组织中细胞凋亡情况,采用Western blot法检测各组大鼠海马组织中p-JAK2、p-STAT3及Bcl-2蛋白表达情况。结果 与假手术组比较,模型组大鼠Morris水迷宫实验第1、2、3、4天逃避潜伏时间均显著延长(P<0.05),穿越平台次数及探索平台速度均显著降低(P<0.05),海马组织细胞凋亡率及海马组织中p-JAK2、p-STAT3、Bcl-2表达水平均显著升高(P<0.05)。与模型组比较,地塞米松组和外源性胆红素低、中、高剂量组大鼠Morris水迷宫实验第1、2、3、4天逃避潜伏时间均显著缩短(P<0.05),穿越平台次数及探索平台速度均显著增加(P<0.05),海马组织细胞凋亡率及海马组织中p-JAK2、p-STAT3、Bcl-2表达水平均显著降低(P<0.05)。与地塞米松组比较,外源性胆红素低、中剂量组大鼠Morris水迷宫实验第1、2、3、4天逃避潜伏时间显著延长(P<0.05),穿越平台次数及探索平台速度均显著降低(P<0.05),海马组织细胞凋亡率及海马组织中p-JAK2、p-STAT3、Bcl-2表达水平均显著升高(P<0.05)。外源性胆红素高剂量组与地塞米松组大鼠Morris水迷宫实验第1、2、3、4天逃避潜伏时间、穿越平台次数及探索平台速度、海马组织细胞凋亡率及海马组织中p-JAK2、p-STAT3、Bcl-2表达水平比较差异均无统计学意义(P>0.05)。随着外源性胆红素剂量增加,外源性胆红素低、中、高剂量组大鼠Morris水迷宫实验第1、2、3、4天大鼠逃避潜伏时间呈缩短趋势,穿越平台次数及探索平台速度呈增加趋势,海马组织细胞凋亡率及海马组织中p-JAK2、p-STAT3、Bcl-2表达水平呈降低趋势,组间两两比较差异均有统计学意义(P<0.05)。结论 外源性胆红素能够提高缺血缺氧性脑损伤大鼠的认知功能,抑制海马组织细胞凋亡,这一作用可能是通过调控JAK2/STAT3/Bcl-2通路的活性而实现。
Abstract:
Objective To observe the effect of exogenous bilirubin on cognitive function and Janus activated kinase-2 (JAK2)/signal transducer and activator 3 (STAT3)/B-cell lymphoma 2 (Bcl-2) pathway in hippocampal tissue of rats with hypoxic-ischemic brain injury.Methods Seventy-two 7-day-old male Wistar rats without specific pathogens were selected and randomly divided into sham operation group,model group,dexamethasone group,low dose exogenous bilirubin group,middle dose exogenous bilirubin group and high dose exogenous bilirubin group,with 12 rats in each group.The rats in each group were intraperitoneal injection of chloral hydrate for anesthesia,the left common carotid artery was separated,the external carotid artery was ligated with silk thread (sham operation group only wore thread without ligature).Four hours after the model prepared,the rats in dexamethasone group was injected intraperitoneally with dexamethasone(10 mg·kg-1);the rats in low,medium,and high dose exogenous bilirubin group were injected intraperitoneally with 5,10,and 15 mg·kg-1 of exogenous bilirubin,respectively;the rats in the sham operation group and the model group were injected intraperitoneally with the same volume of normal saline;all rats were continuous administration for 2 weeks.The cognitive function of rats in each group was detected by water maze test;the apoptosis of hippocampal cells was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling;the expressions of p-JAK2,p-STAT3 and Bcl-2 protein in hippocampus of rats in each group was detected by Western blot.Results The escape latency of rats in the model group was significantly prolonged than that in the sham operation group at 1,2,3,4 days of Morris water maze test (P<0.05).The number of crossing platforms and the speed of exploration platform of rats in the model group were significantly lower than those in the sham operation group at 1,2,3,4 days of Morris water maze test (P<0.05).The apoptotic rate and the expression levels of p-JAK2,p-STAT3 and Bcl-2 protein in hippocampal tissues of rats in the model group were significantly higher than those in the sham operation group at 1,2,3,4 days of Morris water maze test (P<0.05).Compared with the model group,the escape latency of rats in the dexamethasone group and low,medium and high dose exogenous bilirubin group was significantly shortened (P<0.05),the number of crossing platforms and the speed of exploration platform were significantly increased (P<0.05),the apoptotic rate and the expression levels of p-JAK2,p-STAT3 and Bcl-2 protein in hippocampal tissue were significantly decreased at 1,2,3,4 days of Morris water maze test (P<0.05).Compared with the dexamethasone group,the escape latency of rats in the low,medium dose exogenous bilirubin group was significantly prolonged (P<0.05),the number of crossing platforms and the speed of exploration platform were significantly decreased (P<0.05),the apoptotic rate and the expression levels of p-JAK2,p-STAT3 and Bcl-2 protein in hippocampal tissues were significantly increased at 1,2,3,4 days of Morris water maze test (P<0.05).There was no significant difference in the escape latency time,number of crossing platforms,and speed of exploring platform,apoptotic rate and expre-ssion levels of p-JAK2,p-STAT3,Bcl-2 protein in hippocampal tissues between the high dose exogenous bilirubin group and the dexamethasone group at 1,2,3,4 days of Morris water maze test (P>0.05).With the increase of the dose of exogenous bilirubin,the escape latency time of rats in the low,middle,and high dose exogenous bilirubin groups was shortened,the number of crossing platforms and the speed of exploration platform were increased,the apoptotic rate and the expression levels of p-JAK2,p-STAT3 and Bcl-2 protein in hippocampal tissue were decreased at 1,2,3,4 days of Morris water maze test,the difference was statistically significant between the low dose exogenous bilirubin group and middle dose exogenous bilirubin group and high dose erogenous bilirubin group (P< 0.05).Conclusion Exogenous bilirubin can improve cognitive function and inhibit hippocampal cell apoptosis in rats with hypoxic-ischemic brain injury,which may be achieved by regulating the activity of JAK2/STAT3/Bcl-2 pathway.
