摘要:
胶质瘤是中枢神经系统最常见的原发性恶性肿瘤,目前其治疗方案仍然是传统的手术联合放射治疗和化学治疗。由于胶质瘤位置特殊、复发率高以及对现有化学治疗药物的耐药性,患者的中位生存期仅15个月左右,因此,迫切需要新的诊断和治疗方案。分化抑制因子2(ID2)是螺旋-环-螺旋转录因子家族成员,在组织中与Ⅰ类和Ⅱ类碱性螺旋-环-螺旋转录因子、视网膜母细胞瘤蛋白及其相关口袋蛋白等相互作用,协同调节免疫细胞分化和基因表达。ID2表达失调与包括胶质瘤在内的多种肿瘤的生长、新生血管形成、侵袭与转移、化学治疗耐药和肿瘤细胞干性有关。本文从ID2的分子结构、基本功能、表达调控、对胶质瘤进展的影响等方面进行综述,旨在为基于ID2靶点的胶质瘤治疗药物的研发提供新思路。
Abstract:
Glioma is the most common primary malignant tumor of the central nervous system.At present,its treatment strategies are still traditional surgery combined with radiotherapy or chemotherapy.Due to specific location,high recurrence rate and drug resistance to chemotherapy of glioma,the median survival of patients is only about 15 months.Therefore,the new diagnosis and treatment for glioma is needed.Inhibitor of differentiation 2 (ID2),a member of the helix loop helix transcription factor family,can interact with class Ⅰ and classⅡ basic helix loop helix transcription factors,retinoblastoma protein and related pocket proteins in tissues to regulate immune cell differentiation and gene expression synergistically.Dysregulated ID2 expre-ssion is associated with the growth,neovascularization,tumor invasion and metastasis,chemotherapy resistance and tumor cell stemness of a variety of tumors(including gliomas).This paper reviewed the molecular structure,basic functions,expression regulation and its impact on the development of glioma,in order to provide new insights for the research and development of ID2 target-based therapeutic drugs of glioma.
