摘要:
目的 探讨微RNA-143(miR-143)和丝裂原活化蛋白激酶7(MAPK7)在子宫内膜异位症(EMT)患者子宫内膜组织中的表达及意义。方法 选择2018年3月至2019年3月于重庆市九龙坡区中西医结合医院行开腹或腹腔镜手术治疗的35例EMT患者作为观察组,另选择行输卵管吻合术的35例输卵管堵塞患者(非子宫内膜异位症患者)作为对照组。收集观察组患者在位子宫内膜组织、异位子宫内膜组织和对照组患者正常子宫内膜组织标本,采用实时荧光定量聚合酶链反应法检测子宫内膜组织中miR-143和MAPK7 mRNA的表达;采用Pearson相关系数分析EMT患者子宫内膜组织中miR-143与MAPK7 mRNA表达的相关性。结果 观察组患者在位子宫内膜组织、异位子宫内膜组织中miR-143、MAPK7 mRNA相对表达量显著高于对照组(P<0.05),观察组患者异位子宫内膜组织中miR-143、MAPK7 mRNA相对表达量显著高于在位子宫内膜组织(P<0.05)。对照组患者分泌期子宫内膜组织中miR-143、MAPK7 mRNA相对表达量显著高于增生期子宫内膜组织(P<0.05),观察组患者增生期与分泌期在位子宫内膜组织、异位子宫内膜组织中miR-143、MAPK7 mRNA相对表达量比较差异均无统计学意义(P>0.05)。Pearson相关性分析结果显示,EMT患者在位子宫内膜组织、异位子宫内膜组织中miR-143与MAPK7 mRNA表达均呈正相关(r=0.515、0.572,P<0.05)。结论 EMT患者子宫内膜组织中miR-143、MAPK7 mRNA表达水平显著升高,且EMT患者子宫内膜组织中miR-143与MAPK7 mRNA表达呈显著正相关,二者可能参与了EMT的发病过程。
Abstract:
Objective To investigate the significance of the expression of microRNA-143 (miR-143) and mitogen activated protein kinase 7 (MAPK7) in endometrium of patients with endometriosis (EMT).Methods Thirty-five patients with EMT who underwent laparotomy or laparoscopic surgery in Chongqing Jiulongpo District Hospital of Integrated Traditional Chinese and Western Medicine from March 2018 to March 2019 were selected as the observation group,and thirty-five patients with tubal obstruction (non-endometriosis patients) who underwent tubal anastomosis were selected as the control group.The eutopic endometrial tissues,ectopic endometrial tissues of patients in the observation group and the normal endometrial tissues of patients in the control group were collected.The expressions of miR-143 and MAPK7 mRNA in endometrial tissues were detected by real-time fluorescence quantitative polymerase chain reaction.The correlation between miR-143 and MAPK7 mRNA expression in endometrium of EMT patients was analyzed by Pearson correlation analysis.Results The relative expression of miR-143 and MAPK7 mRNA in eutopic endometrium and ectopic endometrium of patients in the observation group were significantly higher than those in the control group (P<0.05).The relative expression of miR-143 and MAPK7 mRNA in ectopic endometrial tissues were significantly higher than those in eutopic endometrial tissues of patients in the observation group (P<0.05).In the control group,the relative expression of miR-143 and MAPK7 mRNA in secretory endometrial tissues were significantly higher than those in proliferative endometrial tissues (P<0.05).In the observation group,there was no significant difference in the relative expression of miR-143 and MAPK7 mRNA in eutopic endometrium and ectopic endometrium between proliferative stage and secretory stage (P>0.05).Pearson correlation analysis showed that there was a positive correlation between miR-143 and MAPK7 mRNA expression in eutopic endometrium and ectopic endometrium of EMT patients (r=0.515,0.572P<0.05).Conclusion The expression levels of miR-143 and MAPK7 mRNA in endometrium of EMT patients were significantly increased,and there was a significant positive correlation between miR-143 and MAPK7 mRNA in endometrium of EMT patients,they may be involved in the pathogenesis of EMT.
