摘要:
目的 探讨藏红花素对人宫颈癌Hela细胞顺铂耐药性的影响。方法 将对数生长期耐顺铂人宫颈癌Hela/DDP细胞随机分为空白对照组、藏红花素组、顺铂组及联合组。藏红花素组细胞以含终质量浓度400 mg·L-1 藏红花素的培养基培养,顺铂组细胞以含终质量浓度5 mg·L-1顺铂的培养基培养,联合组细胞以含终质量浓度400 mg·L-1藏红花素和 5 mg·L-1顺铂的培养基培养,空白对照组细胞以正常培养基培养,每组设10个复孔。培养48 h后,采用细胞计数试剂盒-8检测各组细胞增殖抑制率,流式细胞术检测各组细胞凋亡情况和细胞周期分布,Western blot法检测各组细胞中激活型半胱氨酸蛋白酶-3(cleaved caspase-3)、B淋巴细胞瘤-2(Bcl-2)、Bcl-2相关X蛋白(Bax)、细胞周期蛋白D1(cyclin D1)、周期蛋白依赖性激酶2(CDK2)蛋白的表达。结果 与空白对照组比较,藏红花素组、顺铂组、联合组细胞增殖抑制率、细胞凋亡率、G0/G1期细胞比例及cleaved caspase-3、Bax、Bcl-2蛋白相对表达量、Bax/Bcl-2值升高(P<0.05),g>2/M期细胞比例及cyclin D1、CDK2蛋白相对表达量降低(P<0.01)。藏红花素组细胞增殖抑制率、细胞凋亡率、g>0/G1期细胞比例、G2/M期细胞比例、Bax/Bcl-2值及cleaved caspase-3、Bax、Bcl-2、CDK2蛋白相对表达量与顺铂组比较差异均无统计学意义(P>0.05);与顺铂组比较,藏红花素组细胞中cyclin D1蛋白相对表达量降低(P<0.01)。与顺铂组和藏红花素组比较,联合组细胞增殖抑制率、细胞凋亡率、g>0/G1期细胞比例、Bax/Bcl-2值及cleaved caspase-3、Bax蛋白相对表达量升高(P<0.05),g>2/M期细胞比例及cyclin D1、CDK2蛋白相对表达量降低(P<0.05)。空白对照组、藏红花素组、顺铂组、联合组s期细胞比例比较差异无统计学意义(>P>0.05)。结论 藏红花素能够逆转人宫颈癌细胞对顺铂的耐药性,其机制可能与调节凋亡相关蛋白表达、促进Hela/DDP细胞凋亡、阻滞Hela/DDP细胞周期进程有关。undefinedundefinedundefinedundefinedundefined
Abstract:
Objective To investigate the effect of crocin on the cisplatin resistance of human cervical cancer Hela cells.Methods The cisplatin-resistant Hela/DDP cells of human cervical cancer in logarithmic growth phase were randomly divided into blank control group,crocin group,cisplatin group and combination group;the cells in the crocin group were cultured with medium containing a final concentration of 400 mg·L-1 crocin,the cells in the cisplatin group were cultured with medium containing a final concentration of 5 mg·L-1 cisplatin,the cells in the combination group were cultured with medium containing a final concentration of 400 mg·L-1 crocin and 5 mg·L-1 cisplatin,the cells in the blank control group were cultured with normal culture medium;each group had 10 multiple holes.After 48 h of culture,the cell proliferation inhibition rate of cells in each group was detected by cell counting kit-8.The cell apoptosis and cell cycle distribution of cells in each group were detected by flow cytometry.The relative expression levels of cleaved cysteinyl aspartate specific proteinase-3 (cleaved caspase-3),B-cell lymphoma/leukemia-2 (Bcl-2),Bcl-2 associated X protein (Bax),cyclin D1,cyclin-dependent kinase 2 (CDK2) protein were detected by Western blot method.Results Compared with the blank control group,the cell proliferation inhibition rate,apoptosis rate,G0/G1 phase cell ratio,the Bax/Bcl-2 value and the relative expression levels of cleaved caspase-3,Bax and Bcl-2 protein of cells in the crocin group,cisplatin group and combination group significantly increased (P<0.05);the ratio of G2/M phase cells and the relative expression levels of cyclin D1 and CDK2 protein significantly decreased (P<0.01).There was no significant difference in cell proliferation inhibition rate,apoptosis rate,G0/G1 phase cell ratio,G2/M phase cell ratio,Bax/Bcl-2 value and the relative expression levels of cleaved caspase-3,Bax,Bcl-2,CDK2 protein between the crocin group and the cisplatin group(P>0.05).The relative expression level of cyclin D1 protein of cells in the crocin group was significantly lower than that in the cisplatin group(P<0.01).Compared with the cisplatin group and crocin group,the cell proliferation inhibition rate,apoptosis rate,G0/G1 phase cell ratio,Bax/Bcl-2 value and the relative expression level of cleaved caspase-3,Bax protein of cells in the combination group were higher (P<0.05);the G2/M phase cells ratio and the relative expression levels of cyclin D1 and CDK2 protein were significantly lower (P<0.05).There was no significant difference in the S phase cell ratio of cells among the blank control group,crocin group,cisplatin group and combination group (P﹥0.05).Conclusion Croctin can reverse the drug resistance of human cervical cancer cells to cisplatin,and the mechanism may be related to regulating the expression of apoptosis-related proteins,Promoting apoptosis and blocking cell cycle progression of Hela/DDP cells.
