摘要:
目的 探讨宫颈癌患者癌细胞中长链非编码RNA(lncRNA)SOX21-AS1基因启动子区甲基化水平变化及其对宫颈癌的诊断价值。方法 选择2016年8月至2018年8月在郑州大学第三附属医院接受宫颈液基细胞学检查的患者43例为研究对象。根据检查结果将患者分为癌症组(细胞学和组织学同时诊断为宫颈鳞状细胞癌,n=23)和非癌症组(细胞学诊断正常且人乳头癌病毒检测为阴性,n=20)。采用焦磷酸测序技术检测宫颈癌细胞中lncRNA SOX21-AS1基因启动子区甲基化水平,并应用受试者工作特征曲线评估其对宫颈癌的诊断价值。结果 宫颈癌患者的年龄、肿瘤直径、组织学分级、淋巴结转移及肌层浸润深度与lncRNA SOX21-AS1基因甲基化水平均无关(P>0.05),患者的国际妇产科联盟分期与lncRNA SOX21-AS1基因甲基化水平有关(P<0.05)。宫颈癌组患者lncrna sox21-as1基因启动子区甲基化水平显著低于非癌症组="" (p<0.05)。宫颈癌患者lncrna="" sox21-as1基因启动子区甲基化水平与宫颈癌的发生呈负相关(r="-0.528,P<0.05)。lncRNA" sox21-as1基因启动子区低甲基化诊断宫颈癌的曲线下面积为="" 0.846,敏感度为100.0%,特异度为69.2%。结论 lncRNA SOX21-AS1基因启动子区低甲基化对宫颈癌具有良好的诊断价值。undefined/html>
Abstract:
Objective To investigate the alteration of methylation level in the promoter region of long non-coding RNA (lncRNA) SOX21-AS1 gene in cancer cell and its diagnostic value in cervical cancer.Methods  Forty-three patients who received cervical liquid based cytology examination in the Third Affiliated Hospital of Zhengzhou University from August 2016 to August 2018 were selected as the research objects.The patients were divided into cancer group (both cytology and histology diagnosed the patient as cervical squamous cell carcinoma,n=23) and normal group (cytological diagnosis was normal and the human papiLlomavirus was negative,n=20) according to the examination results.The methylation level in the promoter region of lncRNA SOX21-AS1 gene in cervical cancer cells was detected by pyrosequencing,and the its diagnostic value in cervical cancer was evaluated by receiver operating characteristic curve.Results The age,tumor diameter,histological grade,lymph node metastasis and myometrial invasion depth of patients with cervical cancer were not related to the methylation level of lncRNA SOX21-AS1 gene (P>0.05),but the Federation International of Gynecology and Obstetrics stage was related to the methylation level of lncRNA SOX21-AS1 gene (P<0.05).The methylation level in the promoter region of lncRNA SOX21-AS1 gene of patients in cervical cancer group was significantly lower than that in normal group (P<0.05).The methylation level in the promoter region of lncRNA SOX21-AS1 gene was negatively correlated with the occurrence of cervical cancer (r=-0.528,P<0.05).The area under the curve of hypomethylation in promoter of lncRNA SOX21-AS1 gene in diagnosing cervical cancer was 0.846,the sensitivity was 100.0%,and the specificity was 69.2%.Conclusion Hypomethylation of lncRNA SOX21-AS1 promoter region has good diagnostic value for cervical cancer.
