摘要:
目的 探讨间隔区寡核苷酸DNA微阵列法在结核分枝杆菌基因分型检测中的应用价值。方法 将结核分枝杆菌标准菌株(H37Rv)、卡介苗(BCG)菌株和97株临床结核分枝杆菌株的聚合酶链反应产物分别进行传统Spoligotyping和间隔区寡核苷酸DNA微阵列分型,比较2种方法的一致性,并用H37Rv菌株检测间隔区寡核苷酸DNA微阵列法的灵敏度。结果 结核分枝杆菌标准菌株、BCG菌株和97株临床结核分枝杆菌菌株的间隔区寡核苷酸DNA微阵列分型结果与传统Spoligotyping完全一致,符合率为100%;且间隔区寡核苷酸DNA微阵列法有较好的检测效能。结论 间隔区寡核苷酸 DNA 微阵列法用于结核分枝杆菌分型具有操作简便、快速等特点,适于结核病的溯源和分子流行病学调查。
Abstract:
Objective To investigate the application value of spacer oligonucleotide typing (Spoligotyping) base on DNA microarray genotyping in Mycobacterium tuberculosis.Methods  The polymerase chain reaction products of the standard strains of Mycobacterium tuberculosis,bacille calmette-guerin (BCG) and 97 strains of clinical Mycobacterium tuberculosis were detected with traditional Spoligotyping and DNA microarray method,respectively.The consistency of the two methods was compared,and the sensitivity of DNA microarray genotyping was detected by H37Rv strain.Results The results of spacer oligonucleotide DNA microarray typing of Mycobacterium tuberculosis standard strains,BCG strains and 97 strains of clinical Mycobacterium tuberculosis were completely consistent with that of traditional Spoligotyping,the coincidence rate was 100%,and the detection efficiency of Spoligotyping base on DNA microarray genotyping was better.Conclusion The Spoligotyping base on DNA microarray in the typing of Mycobacterium tuberculosis is simple and rapid,which is suitable for the traceability and molecular epidemiological investigation of tuberculosis.
