摘要:
目的 探讨以胆碱及甜菜碱为主要成分的甲基营养素对大鼠同型半胱氨酸(Hcy)水平的影响。方法 将80只Sprague Dawley大鼠随机分为对照组、模型组、预防组1、预防组2、预防组3、预防组4、治疗组1、治疗组2、治疗组3、治疗组4,每组8只。对照组大鼠正常饲料喂养,模型组大鼠给予含质量分数2%蛋氨酸饲料喂养制备高同型半胱氨酸血症(Hhcy)模型,预防组1大鼠给予含质量分数2%蛋氨酸和叶酸的饲料喂养,预防组2大鼠给予含质量分数2%蛋氨酸、叶酸、维生素B6、维生素B12的饲料喂养,预防组3大鼠给予含质量分数2%蛋氨酸、叶酸、维生素B6、维生素B12、胆碱,预防组4给予叶酸+维生素B6+维生素B12+甜菜碱,共喂养45 d。治疗组1、治疗组2、治疗组3、治疗组4大鼠前30 d给予含质量分数2%蛋氨酸的饲料喂养,之后分别添加与预防组1~4相对应的营养素喂养45 d。对照组、模型组、预防组大鼠于第45天,治疗组大鼠于第75天采用酶联免疫吸附试验测大鼠血清中Hcy水平。结果 模型组大鼠血清中Hcy水平显著高于对照组(P<0.05);预防组1、预防组2、预防组3、预防组4大鼠血清中Hcy水平均低于模型组(P<0.05);预防组2、预防组3、预防组4大鼠血清中Hcy水平均低于预防组1(P<0.05);预防组2、预防组3、预防组4大鼠血清中Hcy水平比较差异均无统计学意义(P>0.05);预防组3、预防组4大鼠血清中Hcy水平与对照组比较差异无统计学意义(P>0.05)。治疗组4大鼠血清中Hcy水平低于模型组(P<0.05),其余各治疗组大鼠血清中Hcy水平与模型组比较差异均无统计学意义(P>0.05)。结论 以胆碱和甜菜碱为主要成分的甲基营养素对HHcy有一定的预防作用,其对HHcy的治疗效果尚不确定。
Abstract:
Objective To investigate the effect of choline and betaine-basedmethyl nutrients on the level of homocysteine in rats.Methods  A total of 80 Sprague Dawley rats were randomly divided into control group,model group,Prevention group 1,Prevention group 2,Prevention group 3,Prevention group 4,treatment group 1,treatment group 2,treatment group 3 and treatment group 4,with 8 rats in each group.The rats in the control group were fed with normal diet,the rats in model group were fed with mass fraction 2% methionine to prepare hyperhomocysteinaemia(HHcy) model,the rats in prevention group 1 were fed with mass fraction 2% methionine and folic acid,the rats in prevention group 2 were fed with mass fraction 2% methionine,folic acid,vitamin B6 and vitamin B12,the rats in prevention group 3 were fed with mass fraction 2% methionine,folic acid,vitamin B6,vitamin B12 and choline,the rats in the prevention group 4 were fed with mass fraction 2% methionine,folic acid,vitamin B6,B12 and betaine;the rats in above six groups were fed for 45 days.The rats in treatment group 1,treatment group 2,treatment group 3 and treatment group 4 were fed with mass fraction 2%methionine for the first 30 days,and then they were fed with nutrients corresponding to prevention group 1-4 for 45 days.On the 45th day in the control group,model group and prevention group,and on the 75th day in the treatment group,the serum levels of homocysteine of rats were measured by enzyme-linked immunosorbent assay.Results The levels of serum Hcy of rats in the model group was significantly higher than that in the control group(P<0.05);the levels of serum Hcy of rats in the prevention group 1,Prevention group 2,Prevention group 3 and prevention group 4 were significantly lower than those in the model group (P<0.05);the levels of serum Hcy of rats in the prevention group 2,Prevention group 3 and prevention group 4 were significantly lower than those in the prevention group 1 (P<0.05);there was no statistical difference in the levels of serum Hcy of rats among prevention group 2,Prevention group 3 and prevention group 4 (P>0.05);there was no statistical difference in the levels of serum Hcy of rats between the prevention group 3,Prevention group 4 and control group(P>0.05).The serum level of Hcy of rats in the treatment group 4 was significantly lower than that in the model group (P<0.05);and there was no significant difference in the levels of serum Hcy of rats between the other treatment groups and model group (P>0.05).Conclusion The choline and betaine-based methyl nutrients have a certain preventive effect on HHcy,but its therapeutic effect on HHcy is still uncertain.
