摘要:
目的 探讨大黄酸对白血病细胞脂肪量和肥胖相关蛋白(FTO)表达及白血病细胞增殖和凋亡的影响及其可能的分子机制。方法 将对数生长期白血病细胞HL60、NB4、Naml6、THP1细胞随机分为对照组及10、20、40、80 μmol·L-1大黄酸组,分别用终浓度0、10、20、40、80 μmol·L-1的大黄酸处理后培养;于培养24、48、72 h 时,采用细胞计数试剂盒检测细胞的增殖能力;于培养72 h时,使用流式细胞仪检测各组细胞的凋亡情况,Western blot检测各组细胞中FTO和caspase-3蛋白相对表达量。结果 培养24、48、72 h时,各浓度大黄酸组HL60、NB4、Nalm6细胞增殖抑制率均高于对照组(P<0.05),随大黄酸浓度增加,hl60、nb4、nalm6细胞增殖抑制率显著增高,呈浓度依赖性(>P<0.05)。10、20、40、80 μmol·l-1大黄酸组THP1细胞增殖抑制率高于对照组(P<0.05)10、20、40、80 μmol·l-1大黄酸组THP1细胞增殖抑制率比较差异均无统计学意义(P>0.05)。各浓度大黄酸组HL60、NB4、Nalm6细胞凋亡率均显著高于对照组(P<0.05);随大黄酸浓度增加,hl60、nb4细胞凋亡率显著增加,呈浓度依赖性(>P<0.05);40、80 μmol·l-1大黄酸组Nalm6细胞凋亡率显著高于10 μmol·L-1大黄酸组和20 μmol·L-1大黄酸组(P<0.05);对照组、10 μmol·l-1大黄酸组和20 μmol·L-1大黄酸组Nalm6细胞凋亡率比较差异无统计学意义(P>0.05)。各浓度大黄酸组HL60、NB4、Nalm6细胞中FTO蛋白相对表达量显著低于对照组,caspase-3蛋白相对表达量显著高于对照组(P<0.05);随大黄酸浓度增加,hl60、nb4、nalm6细胞中fto蛋白相对表达量显著降低,caspase-3蛋白相对表达量显著增高,呈浓度依赖性(>P<0.05)。5组thp1细胞凋亡率及thp1细胞中fto和caspase-3蛋白相对表达量比较差异均无统计学意义(>P>0.05)。结论 大黄酸可通过抑制FTO的表达,抑制白血病细胞的增殖并促进其凋亡,且对HL60和NB4细胞的作用明显强于Naml6和THP1细胞。undefinedundefinedundefinedundefinedundefinedundefinedundefinedundefined/sup="">/sup="">
Abstract:
Objective To investigate the effect of rhein on the expression of fat mass and obesity-associated protein(FTO) and proliferation,apoptosis of leukemia cells,and its possible molecular mechanisms.Methods  The leukemia cells HL60,NB4,Naml6 and THP1 were randomly divided into control group and 10,20,40,80 μmol·L-1 rhein groups.The cells in the control group and 10,20,40,80 μmol·L-1 rhein groups were treated with the final concentration of 0,10,20,40 and 80 μmol·L-1 rhein,respectively.At 24,48,72 hours of culture,cell counting kit-8 was used to determine the proliferation capacity of cells in every group.At 72 hours of culture,flow cytometry was conducted to detect the cell apoptosis,Western blot was used to detect the relative expression levels of FTO and caspase-3 protein.Results At 24,48,72 h of culture,the proliferation inhibition rate of HL60,NB4 and Nalm6 cells in every concentration of rhein groups were significantly higher than that in the control group (P<0.05),with the proliferation increase of rhein concentration,the proliferation inhibition rate of HL60,NB4 and Nalm6 cells increased significantly in a concentration dependent manner (P<0.05).The proliferation inhibition rate of THP1 cells in 10,20,40 and 80 μmol · L-1 rhein groups was higher than that in the control group (P<0.05);there was no significant difference in the proliferation inhibition rate of THP1 cells in 10,20,40 and 80 μmol · L-1 rhein groups (P>0.05).The apoptosis rate of HL60,NB4 and Nalm6 cells in every concentration of rhein groups were significantly higher than that in the control group (P<0.05);with the increase of rhein concentration,the apoptosis rate of HL60 and NB4 cells increased significantly in a concentration dependent manner (P<0.05);the apoptosis rate of Nalm6 cells in 40,80 μmol · L-1 rhein groups was significantly higher than that in 10 μmol · L-1 rhein group and 20 μmol · L-1 rhein group (P<0.05);there was no significant difference in the apoptosis rate of Nalm 6 cells in the control group,10 μmol · L-1 rhein group and 20 μmol · L-1 rhein group (P>0.05).The relative expression level of FTO protein of HL60,NB4 and Nalm6 cells in every concentration of rhein groups was significantly lower than that in the control group,the relative expression level of caspase-3 protein was significantly higher than that in the control group (P<0.05);with the increase of rhein concentration,the relative expression level of FTO protein in HL60,NB4 and Nalm6 cells decreased significantly,while the relative expression level of caspase-3 protein increased significantly,which were in a concentration dependent manner (P<0.05).There was no significant difference in the apoptosis rate and the relative expression of FTO and caspase-3 protein of THP1 cells in the five groups (P>0.05).Conclusion Rhein can inhibit the proliferation and promote the apoptosis of leukemia cells by inhibiting the FTO expression,these effects of rhein on HL60 and NB4 cells are stronger than those of Nalm6 and THP1 cells.
