摘要:
观察腺苷激酶抑制剂ABT-702对大鼠局灶性脑缺血再灌注损伤早期氧化应激和神经元凋亡的影响。方法 将60只健康Sprague Dawley雄性大鼠随机分为假手术组、缺血再灌注(IR)组和ABT-702干预组,其中IR组和ABT-702干预组大鼠按照恢复灌注的时间点分为IR 2 h组、IR 6 h组和ABT-702 2 h组、ABT-702 6 h组,每组12只。IR组和ABT-702干预组大鼠参照改良的Zea-longa线栓法制备大脑中动脉栓塞模型,假手术组大鼠仅分离颈总动脉、颈外动脉、颈内动脉,不插入线栓。IR组大鼠于恢复灌注时腹腔注射二甲基亚砜稀释液(3 mL·kg-1), ABT-702干预组大鼠于恢复灌注时腹腔注射ABT-702(1.5 mg·kg-1),假手术组大鼠不给予任何干预措施。采用酶联免疫吸附测定法检测脑缺血半暗带组织中超氧化物歧化酶(SOD)活性、脂质过氧化物产物丙二醛(MDA)水平及凋亡因子caspase-3活性。结果 IR 2 h组、IR 6 h组、ABT-702 2 h组、ABT-702 6 h组大鼠脑组织中SOD活性均低于假手术组(P<0.05); ABT-702 2 h组大鼠脑组织中SOD活性显著高于IR 2 h组(P<0.05);ABT-702 6 h组大鼠脑组织SOD活性显著高于IR 6 h组(P<0.05)。IR 2 h组、IR 6 h组、ABT-702 2 h组、ABT-702 6 h组大鼠脑组织中MDA含量均高于假手术组(P<0.05);ABT-702 2 h组大鼠脑组织中MDA含量显著低于IR 2 h组(P<0.05);ABT-702 6 h组大鼠脑组织中MDA含量显著低于IR 6 h组(P<0.05)。IR 2 h组、IR 6 h组、ABT-702 2 h组、ABT-702 6 h组大鼠脑组织中caspase-3活性均高于假手术组;ABT-702 2 h组和IR 2 h组大鼠脑组织中caspase-3活性比较差异无统计学意义(P>0.05);ABT-702 6 h组大鼠脑组织中caspase-3活性显著低于IR 6 h组(P<0.05)。结论 ABT-702在大鼠局灶性脑缺血再灌注损伤早期可能通过抑制自由基的产生而发挥抗氧化作用,同时能减轻缺血半暗带区神经元凋亡,从而对脑组织缺血再灌注损伤早期起保护作用。
Abstract:
Objective To observe the effect of adenosine kinase inhibitor ABT-702 on oxidative stress and neuronal apoptosis in the early stage of focal cerebral ischemia-reperfusion injury in rats.Methods Sixty healthy Sprague Dawley male rats were randomly divided into the sham operation group,ischemia-reperfusion(IR) group,ABT-702 intervention group;the rats in the IR group and the ABT-702 intervention group were divided into two subgroups of IR 2 h group,IR 6 h group and ABT-702 2 h group,ABT-702 6 h group respectively according to the time point of reperfusion,with 12 rats in each group.The rats in the IR group and the ABT-702 intervention group were made the middle cerebral artery occlusion model by modified Zea longa suture method,and the rats in the sham operation group were only isolated the common carotid artery,external carotid artery and internal carotid artery without inserting thread plug.The rats in the IR group were intraperitoneally injected with dimethyl sulfoxide(3 mL·kg-1)at the time of resuming perfusion;the rats in the ABT-702 intervention group were intraperitoneally injected with ABT-702(1.5 mg·kg-1);the rats in the sham operation group were not given any intervention.The activity of superoxide dismutase (SOD),the level of malondialdehyde (MDA) in the lipid peroxidation product and the activity of apoptosis factor caspase-3 in the ischemic penumbra were detected by enzyme linked immunosorbent assay.Results The activity of SOD in brain tissues of rats in the IR 2 h group,IR 6 h group,ABT-702 2 h group and ABT-702 6 h group was lower than that in the sham operation group (P<0.05);the activity of SOD in brain tissues of rats in the ABT-702 2 h group was higher than that in the IR 2 h group (P<0.05);the activity of SOD in brain tissue of rats in the ABT-702 6 h group was higher than that in the IR 6 h group (P<0.05).The level of MDA in brain tissues of rats in the IR 2 h group,IR 6 h group,ABT-702 2 h group and ABT-702 6 h group was higher than that in the sham operation group (P<0.05);the level of MDA in brain tissues of rats in the ABT-702 2 h group was lower than that in the IR 2 h group (P<0.05);the content of MDA in brain tissue of rats in the ABT-702 6 h group was lower than that in the IR 6 h group (P<0.05).The activity of caspase-3 in brain tissues of rats in the IR 2 h group,IR 6 h group,ABT-702 2 h group and ABT-702 6 h group was higher than that in the sham operation group (P<0.05);there was no statistical difference in the activity of caspase-3 in brain tissues of rats between the ABT-702 6 h group and the IR 6 h group(P>0.05);the activity of caspase-3 in brain tissues of rats in the ABT-702 6 h group was lower than that in the IR 6 h group (P<0.05).Conclusion ABT-702 may play an antioxidant role in the early stage of focal cerebral IR injury of rats by inhibiting the production of free radicals,and at the same time can reduce the apoptosis of neurons in the ischemic penumbra area,thus playing a protective role in the early stage of cerebral IR injury.
