摘要:
目的 探讨丙泊酚对创伤后应激障碍(PTSD)大鼠恐惧记忆的影响及可能机制。方法 将60只雄性Sprague Dawley大鼠随机分为对照组、消退对照组、消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组,每组10只。对照组大鼠进行标准饲养,不给予任何处理;其余5组大鼠采用单一连续应激法建立PTSD模型。造模成功后第9~14 天,消退对照组大鼠不给予任何处理,消退训练+丙泊酚低剂量组大鼠每日给予消退训练并腹腔注射0.2 mg·kg-1丙泊酚,消退训练+丙泊酚中剂量组大鼠每日给予消退训练并腹腔注射0.6 mg·kg-1丙泊酚,消退训练+丙泊酚高剂量组大鼠每日给予消退训练并腹腔注射1.0 mg·kg-1丙泊酚,消退训练组大鼠每日给予消退训练并腹腔注射等量生理盐水,连续6 d。采用高架十字迷宫实验测各组大鼠焦虑状态,条件性恐惧记忆实验检测各组大鼠恐惧记忆,酶联免疫吸附试验检测各组大鼠血清皮质酮水平,Western blotting法检测各组大鼠海马组织中脑源性神经营养因子(BDNF)及其特异性受体TrkB蛋白的表达水平。结果 消退对照组、消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠僵直停留时间均显著长于对照组(P<0.05);消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠僵直停留时间均显著短于消退对照组(P<0.05);消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠僵直停留时间均显著短于消退训练组(P<0.05);不同剂量丙泊酚组大鼠僵直停留时间随丙泊酚剂量的增加逐渐缩短,两两比较差异均有统计学意义(P<0.05)。消退对照组、消退训练组、消退训练+丙泊酚低剂量组大鼠开臂停留时间和开臂进入次数显著低于对照组(P<0.05);消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠开臂停留时间和开臂进入次数与对照组比较差异无统计学意义(P>0.05)。消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠开臂停留时间和开臂进入次数均显著高于消退对照组(P<0.05);消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠开臂停留时间和开臂进入次数均显著高于消退训练组(P<0.05);不同剂量丙泊酚组大鼠开臂停留时间和开臂进入次数随丙泊酚剂量的增加逐渐升高,两两比较差异均有统计学意义(P<0.05)。消退对照组、消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠血清皮质酮水平均显著高于对照组(P<0.05);消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠血清皮质酮水平均显著低于消退对照组(P<0.05);消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠血清皮质酮水平均显著低于消退训练组(P<0.05);不同剂量丙泊酚组大鼠血清皮质酮水平随着丙泊酚剂量增加而逐渐降低,两两比较差异均有统计学意义(P<0.05)。消退对照组、消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组大鼠海马组织中BNDF、TrkB蛋白表达水平显著低于对照组(P<0.05);消退训练+丙泊酚高剂量组大鼠海马组织中BNDF、TrkB蛋白表达水平与对照组比较差异无统计学意义(P>0.05)。消退训练组、消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠海马组织中BNDF、TrkB蛋白表达水平均显著高于消退对照组(P<0.05);消退训练+丙泊酚低剂量组、消退训练+丙泊酚中剂量组和消退训练+丙泊酚高剂量组大鼠海马组织中BNDF、TrkB蛋白表达水平均显著高于消退训练组(P<0.05);不同剂量丙泊酚组大鼠海马组织中BNDF、TrkB蛋白表达水平随丙泊酚剂量的增加逐渐升高,两两比较差异均有统计学意义(P<0.05)。结论 丙泊酚有助于促进PTSD大鼠恐惧记忆的消除,改善其焦虑状态。
Abstract:
Objective To investigate the effect of propofol on fear memory of post-traumatic stress disorder (PTSD) rats and its possible mechanism.Methods Sixty male Sprague Dawley rats were randomly divided into control group,regression control group,regression training group,regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group,with 10 rats in each group.The rats in the control group were fed with standard diet without any treatment,and the rats in the other five groups were made the PTSD model by single continuous stress method.At the 9-14 days after successful modeling,the rats in the regression control group were not given any treatment;the rats in the regression training + low dose propofol group were given regression training and intraperitoneal injection of 0.2 mg · kg-1 propofol every day for 6 days;the rats in the regression training + medium dose propofol group were given regression training and intraperitoneal injection of 0.6 mg · kg-1 every day for 6 days;the rats in the regression training + high dose propofol group were given regression training and intraperitoneal injection of 1.0 mg · kg-1 propofol every day for 6 days;the rats in the regression training group were given regression training and intraperitoneal injection of the same amount of normal saline for 6 days.The anxiety state of all rats was measured by elevated plus-maze test;the fear memory of all rats was measured by conditional fear memory test.Serum corticosterone levels of all rats were measured by enzyme linked immunosorbent assay.The expressions of brain-derived neurotrophic factor (BDNF) and its specific receptor TrkB protein were detected by Western blotting.Results The stiffness stay time of rats in the regression control group,regression training group,regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group were significantly longer than those in the control group(P<0.05).The open arm stay time and open arm entry times of rats in the regression training group,regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group were significantly higher than those in the regression control group(P<0.05);the open arm stay time and open arm entry times of rats in the regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group were significantly higher than those in the regression training group(P<0.05).The open arm stay time and open arm entry times of rats in different doses of propofol group gradually increased with the increase of the dosage of propofol (P<0.05).there was no significant difference in the expressions of BNDF,TrkB protein in hippocampus tissue of rats between the regression training + high dose propofol group and the control group(P>0.05).The expressions of BNDF,TrkB protein in hippocampus tissue of rats in the regression training group,regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group were significantly higher than those in the regression control group(P<0.05);the expressions of BNDF,TrkB protein in hippocampus tissue of rats in the regression training + low dose propofol group,regression training + medium dose propofol group and regression training + high dose propofol group were significantly higher than those in the regression training group(P<0.05);the expressions of BNDF,TrkB protein in hippocampus tissue of rats in different doses of propofol group gradually increased with the increase of the dosage of propofol (P<0.05).Conclusion Propofol can promote the elimination of fear memory and improve the anxiety state of PTSD rats.
