摘要:
目的 探讨支气管哮喘患儿外周血单核细胞(PBMC)培养上清液中肿瘤坏死因子-α(TNF-α)、白细胞介素-17(IL-17)、免疫球蛋白E(IgE)水平与PBMC中Toll样受体4(TLR4) mRNA表达的相关性。方法 选择2017年4月至2019年5月中国人民解放军空军军医大学第二附属医院收治的160例支气管哮喘患儿,其中急性发作期患儿91例(急性发作组),缓解期患儿69例(缓解组),另选择同期体检健康儿童160例为健康对照组。采用双抗体夹心法酶联免疫吸附试验测定PBMC培养上清液中TNF-α、IL-17、IgE水平,实时荧光定量聚合酶链反应检测PBMC中TLR4 mRNA表达。结果 急性发作组患儿PBMC培养上清液中TNF-α、IL-17、IgE水平高于缓解组和健康对照组,缓解组患儿PBMC培养上清液中TNF-α、IL-17、IgE水平高于健康对照组(P<0.05)。急性发作组TLR4 mRNA相对表达量高于缓解组及健康对照组,缓解组患儿PBMC中TLR4 mRNA相对表达量高于健康对照组(P<0.05)。急性发作组TLR4阻断后PBMC培养上清液中TNF-α、IL-17、IgE水平均低于阻断前(P<0.05)。支气管哮喘患儿PBMC培养上清液中TNF-α、IL-17、IgE水平与PBMC中TLR4 mRNA表达呈正相关(r=6.272、7.653、7.019,P<0.05)。结论 支气管哮喘患儿PBMC培养上清液中TNF-α、IL-17、IgE表达升高,且与PBMC中TLR4表达呈正相关,TLR4可能通过调控TNF-α、IL-17、IgE的表达而参与支气管哮喘的发生和发展。
Abstract:
Objective To investigate the correlation between tumor necrosis factor-α(TNF-α),interleukin-17(IL-17),immunoglobulin E(IgE) levels and the expression of Toll-like receptor 4(TLR4) in peripheral blood mononuclear cell(PBMC) culture supernatants of children with bronchial asthma.Methods A total of 160 children with bronchial asthma admitted to the Second Affiliated Hospital of Chinese PLA Air Force Medical University from April 2017 to May 2019 were selected as the research objects,including 91 children in acute attack stage (acute attack group) and 69 children in remission stage (remission group).Another 160 healthy children in the same period were selected as the healthy control group.The levels of TNF-α,IL-17 and IgE in the supernatant of PBMC culture solution were detected by enzyme-linked immunosorbent assay with double antibody sandwich method;and the expression of TLR4 mRNA in PBMC was detected by real-time quantitative polymerase chain reaction.Results The levels of TNF-α,IL-17 and IgE in PBMC culture supernatant in the acute attack group were higher than those in the remission group and healthy control group;and the levels of TNF-α,IL-17 and IgE in PBMC culture supernatant in the remission group were higher than those in the healthy control group(P<0.05).The relative expression of TLR4 mRNA in PBMC in the acute attack group was higher than that in the remission group and healthy control group (P<0.05),and the relative expression of TLR4 mRNA in PBMC in the remission group was higher than that in the healthy control group (P<0.05).The levels of TNF-α,IL-17 and IgE in PBMC culture supernatant after TLR4 blocking were lower than those before blocking in the acute attack group(P<0.05).The levels of TNF-α,IL-17 and IgE in PBMC culture supernatant were positively correlated with TLR4 mRNA expression in PBMC (r=6.272,7.653,7.019;P<0.05).Conclusion The expression of TNF-α,IL-17 and IgE in PBMC culture supernatant increases in children with bronchial asthma,and they are positively correlated with the expression of TLR4 in PBMC.TLR4 may be involved in the occurrence and development of bronchial asthma by regulating the expression of TNF-α,IL-17 and IgE.
