摘要:
目的 探讨microRNA-182(miR-182)、Ki-67在Ⅰ型和Ⅱ型子宫内膜癌(EC)组织中的表达。方法 选择2017年11月至2019年4月新乡医学院第一附属医院妇科保存的EC组织标本68例(Ⅰ型53例,Ⅱ型15例)和正常子宫内膜组织70例,采用免疫组织化学法检测正常子宫内膜组织和EC组织中雌激素受体(ER)、孕激素受体(PR)和Ki-67的表达,实时荧光定量聚合酶链反应检测正常子宫内膜和EC组织中miR-182相对表达量,并分析Ki-67、miR-182表达与EC临床病理特征的关系。结果 Ⅰ、Ⅱ型EC组织及正常子宫内膜组织中ER阳性表达率分别为98.11(52/53)、13.33%(2/15)、100.0%(70/70),PR阳性表达率分别为100.00%(53/53)、13.33%(2/15)、100.00%(70/70);Ⅰ型EC组织与正常子宫内膜组织中ER阳性表达率比较差异无统计学意义(P>0.05),Ⅱ型EC组织中ER、PR阳性表达率显著低于Ⅰ型EC组织和正常子宫内膜组织(P<0.05);Ⅰ、Ⅱ型EC组织及正常子宫内膜组织中Ki-67阳性表达率分别为81.13%(43/53)、100.00%(15/15)、20.00%(14/70),Ⅰ、Ⅱ型EC组织中Ki-67阳性表达率显著高于正常子宫内膜组织(P<0.05),Ⅱ型EC组织中Ki-67阳性表达率显著高于Ⅰ型EC组织(P<0.05)。EC组织中Ki-67表达与EC类型、EC细胞组织学分级、临床分期、ER、PR表达有关(P<0.05),与患者年龄无关(P>0.05)。正常子宫内膜组织及Ⅰ、Ⅱ型EC组织中miR-182相对表达量分别为 0.133±0.031、1.413±1.012、4.831±1.523,Ⅰ、Ⅱ型EC组织中miR-182相对表达量显著高于正常子宫内膜组织(P<0.05),Ⅱ型EC组织中miR-182相对表达量显著高于Ⅰ型EC组织(P<0.05);miR-182表达与EC类型、EC临床分期及ER、PR表达有关(P<0.05),与患者年龄、EC组织学分级及Ki-67表达无关(P>0.05)。结论 EC组织中miR-182、Ki-67表达上调,Ⅱ型EC组织中miR-182、Ki-67表达高于Ⅰ型EC,miR-182、Ki-67高表达提示患者预后不良。
Abstract:
Objective To investigate the expression of microRNA-182 (miR-182) and Ki-67 in type Ⅰ and type Ⅱ endometrial carcinoma (EC).Methods Sixty-eight tissue samples (53 cases of type Ⅰ EC,15 cases of type Ⅱ EC) and 70 normal endometrial tissue samples in the First Affiliated Hospital of Xinxiang Medical College from November 2017 to April 2019 were collected.The expression of estrogen receptor (ER),progesterone receptor (PR) and Ki-67 in normal endometrium and EC tissues were detected by immunohistochemistry.The expression of miR-182 in normal endometrium and EC tissues were detected by real-time fluorescence quantitative polymerase chain reaction.The relationship between the expression of Ki-67 and miR-182 and the clinicopathological characteristics of EC was analyzed.Results The positive expression rate of ER in type Ⅰ and Ⅱ EC and normal endometrium tissues was 98.11% (52/53),13.33% (2/15) and 100.0% (70/70),respectively.The positive expression rate of PR in type Ⅰ and Ⅱ EC and normal endometrium tissues was 100.00% (53/53),13.33% (2/15) and 100.0% (70/70),respectively.There was no signifcant difference in the positive expression rate of ER and PR between type Ⅰ EC tissues and normal endometrium tissues (P>0.05).The positive expression rate of ER and PR in type Ⅱ EC tissues was significantly lower than that in type Ⅰ EC and normal endometrium tissues (P<0.05).The positive expression rate of Ki-67 in type Ⅰ and Ⅱ EC and normal endometrium tissues was 81.13% (43/53),100.00% (15/15) and 20.00% (14/70),respectively.The positive expression rate of Ki-67 in type Ⅰ and Ⅱ EC tissues was significantly higher than that in normal endometrium (P<0.05).The positive expression rate of Ki-67 in type Ⅱ EC tissues was significantly higher than that in type Ⅰ EC tissues (P<0.05).The expression of Ki-67 in EC tissues was related to the cell histological classification,clinical stage,ER and PR expression (P<0.05),but it was not related to the age of patients (P>0.05).The relative expression of miR-182 in normal endometrium and type Ⅰ and Ⅱ EC tissues was 0.133±0.031,1.413±1.012 and 4.831±1.523,respectively.The relative expression of miR-182 in type Ⅰ and Ⅱ EC tissues was significantly higher than that in normal endometrium tissues (P<0.05).The relative expression of miR-182 in type Ⅱ EC tissues was significantly higher than that in type Ⅰ EC tissues (P<0.05).The expression of miR-182 was related to the type of EC,the clinical stage of EC and the expression of ER and PR (P<0.05),but it was not related to the age of patients,the histological grade of EC and the expression of Ki-67 (P>0.05).Conclusions The expression of miR-182 and Ki-67 in EC tissues wis up-regulated,and the expression of miR-182 and Ki-67 in type Ⅱ EC is higher than that in type Ⅰ EC.The high expression of miR-182 and Ki-67 suggests the unfavourable prognosis of the patients.
