摘要:
目的 分析人中分化肺腺癌及癌旁正常组织中的差异蛋白,寻找与肺腺癌发病相关的蛋白质。方法 利用双向凝胶电泳法分离肺腺癌及癌旁正常组织中的蛋白质,通过图像扫描寻找差异2倍以上蛋白点,进而应用基质辅助激光电离飞行时间质谱得到肽质量指纹图谱,对比数据库确定差异蛋白。结果 肺腺癌组织及癌旁正常组织平均蛋白点数分别为1 810±167、1 704±53。共筛选出18个差异蛋白点,其中13个差异蛋白点在肺腺癌组织中表达上升。结论 双向凝胶电泳技术结合基质辅助激光电离飞行时间质谱可鉴定肺腺癌及癌旁正常组织的差异蛋白,为早期诊断肺癌、判断预后提供依据。
Abstract:
Objective To analyze the differential protein of human moderately differentiated lung adenocarcinoma and adjacent tissues,and find the protein related to the pathogenesis of lung adenocarcinoma.Methods The protein in lung adenocarcinoma and adjacent normal tissues were separated by using two dimensional gel electrophoresis(2-DE),and the differential protein spots with a difference of more than two times were identified by image scanning.Then the peptide mass fingerprins of different protein were obtained by using matrix assisted laser ionization time of flight mass spectrometry,and the differential proteins were determined by comparing the databases.Results The average number of protein points in lung adenocarcinoma tissues and adjacent normal tissues were 1 810±167 and 1 704±53,respectively.A total of 18 differentially expressed protein spots were screened out,of which 13 differentially expressed protein spots increased in lung adenocarcinoma tissues.Conclusion 2-DE combined with matrix assisted laser ionization time of flight mass spectrometry can identify differential proteins in lung adenocarcinoma and adjacent normal tissues,and provide evidence for early diagnosis and prognosis of lung adenocarcinoma.
