摘要:
目的 探讨宫颈癌组织中性别基因框蛋白9 (SOX9)的表达及其临床意义。方法 选择2014年9月至2018年2月南京医科大学附属无锡妇幼保健院收治并经术后病理检查确诊的70例宫颈癌患者为宫颈癌组;并选择同期宫颈癌前病变患者70例为癌前病变组,其中宫颈上皮内瘤变(CIN)Ⅰ级30例,CINⅡ级20例,CIN Ⅲ级20例;另选择同期因子宫肌瘤、异常子宫出血、子宫腺肌症等妇科良性疾病行子宫全切者50例为对照组。采用免疫组织化学法观察各组患者宫颈组织中SOX9蛋白表达情况,分析宫颈癌组织中SOX9表达与临床病理特征的关系。结果 宫颈癌组、癌前病变组、对照组患者宫颈组织中SOX9蛋白阳性表达率分别为 77.1%(54/70)、32.9%(23/70)、8.0%(4/50),宫颈癌组患者宫颈组织中SOX9蛋白阳性表达率显著高于癌前病变组和对照组(χ2=27.734、55.837,P<0.05),癌前病变组患者宫颈组织中SOX9蛋白阳性表达率显著高于对照组(χ2=10.335,P<0.05)。癌前病变组患者中,CINⅠ、Ⅱ、Ⅲ级患者宫颈组织中SOX9蛋白阳性表达率分别为13.3%(4/30)、30.0%(6/20)、65.0%(13/20),CIN Ⅱ、Ⅲ级患者宫颈组织中SOX9蛋白阳性表达率显著高于CIN Ⅰ级患者(χ2=2.083、14.275,P<0.05),CIN Ⅲ级患者宫颈组织中SOX9蛋白阳性表达率显著高于CIN Ⅱ级患者(χ2=4.912,P<0.05)。宫颈癌组织中SOX9蛋白表达与临床分期、组织学分级、淋巴结转移有关(P<0.05),与年龄、体质量指数、病理类型无关 (P<0.05)。结论 SOX9在宫颈癌组织中呈高表达,且与肿瘤的临床分期、转移与分化程度密切相关,在宫颈癌的发生、发展过程中起一定作用。
Abstract:
Objective To investigate the expression and clinical significance of sex determining region Y box protein 9 (SOX9) in cervical cancer tissue.Methods Seventy patients with cervical cancer confirmed by postoperative pathology in Wuxi Maternal and Child Health Hospital Affiliated to Nanjing Medical University from September 2014 to February 2018 were selected as cervical cancer group;seventy patients with cervical precancerous lesions during the same period were selected as precancerous lesions group,including 30 cases of cervical intraepithelial neoplasia (CIN) grade I,20 cases of CIN grade II and 20 cases of CIN grade Ⅲ;fifty cases of benign gynecological diseases such as uterine fibroids,abnormal uterine bleeding and adenomyosis were selected as the control group.The expression of SOX9 in cervical tissues of all subjects was observed by immunohistochemistry.The relationship between SOX9 expression and clinicopathological features was analyzed.Results The positive expression rates of SOX9 in the cervical cancer group,precancerous lesions group and control group were 77.1% (54/70),32.9% (23/70) and 8.0% (4/50),respectively.The positive expression rate of SOX9 in cervical tissue of patients in cervical cancer group was significantly higher than that in the precancerous lesion group and control group (χ2=27.734,55.837;P<0.05);the positive expression rate of SOX9 in cervical tissue of patients in precancerous lesion group was significantly higher than that in the control group (χ2=10.335,P< 0.05).The positive expression rates of SOX9 in cervical tissues of patients with CIN grade Ⅰ,Ⅱ and Ⅲ were 13.3% (4/30),30.0% (6/20) and 65.0% (13/20),respectively.The positive expression rates of SOX9 in cervical tissues of patients with CIN grade Ⅱ and Ⅲ were significantly higher than those of patients with grade Ⅰ (χ2=2.083,14.275;P<0.05).The positive expression rate of SOX9 in cervical tissues of patients with CIN grade Ⅲ was significantly higher than that of patients with grade Ⅱ(χ2=4.912,P<0.05).The expression of SOX9 in cervical cancer tissue was correlated with clinical stage,histological grade and lymph node metastasis (P<0.05),but not correlated with age,body mass index and pathological type (P<0.05).Conclusion The expression of SOX9 in cervical cancer tissues is higher.The expression of SOX9 is closely related to the clinical stage,metastasis and differentiation of cervical cancer,and it plays a role in the occurrence and development of cervical cancer.
