摘要:
目的 探讨射频消融(RFA)术对大鼠肝组织中M2型巨噬细胞极化的调控作用。方法 将42只大鼠随机分为RFA组(n=36)和对照组(n=6),RFA组大鼠根据处死时间再分为12、24、72、120、168、336 h组,每组6只。RFA组大鼠给予RFA术建立肝损伤炎症模型,对照组大鼠只连接射频消融设备,不接通电源。12、24、72、120、168 h组大鼠分别于RFA术后12、24、72、120、168 h脱臼处死,对照组大鼠于168 h时全部处死;采用酶联免疫吸附试验检测各组大鼠血清肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)水平;采用7170A全自动生物化学分析仪检测各组大鼠血清丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)水平。336 h组大鼠于RFA术后336 h处死,采用组织免疫荧光法检测各组大鼠肝组织中M2型巨噬细胞阳性表达。结果 成功建立以针道为中心呈坏死区、交界区和炎症充血区的同心圆样的大鼠肝损伤炎症模型,且随着时间的延长,肝组织炎症逐渐减轻,纤维化修复形成。12、24、72、120、168 h组大鼠血清ALT、AST、TNF-α及IL-10水平均高于对照组(P<0.05);24、72、120 h组大鼠ALT、AST、TNF-α及IL-10水平均高于12 h组(P<0.05);168 h组大鼠血清AST水平高于12 h组,TNF-α水平低于12 h组(P<0.05);168 h组与12 h组大鼠血清ALT和IL-10水平比较差异无统计学意义(P>0.05)。168 h组大鼠血清ALT、AST、TNF-α及IL-10水平均低于120 h组(P<0.05);24、72、120 h组大鼠血清ALT、AST、TNF-α及IL-10水平组间两两比较差异均无统计学意义(P>0.05)。对照组及12、24、72、120、168、336 h组大鼠肝组织中M2型巨噬细胞阳性表达率分别为(9.5±0.4)%、(10.1±0.8)%、(10.9±0.5)%、(11.1±1.5)%、(18.7±0.8)%、(24.5±1.8)%和(6.8±0.4)%。168 h组M2型巨噬细胞阳性表达率高于对照组和12、24、72、120、336 h组(χ2=14.91、14.31、13.60、13.36、12.71、17.63,P<0.05)。结论 RFA术可诱导大鼠肝脏巨噬细胞极化为M2型,且其阳性表达率在RFA术后168 h最高。
Abstract:
Objective To investigate the regulation of radiofrequency ablation(RFA) on polarization of type M2 macrophage in hepatic tissue of rats.Methods Forty-two rats were randomly divided into control group(n=6) and RFA group(n=36);the rats in RFA group were divided into 12 h group,24 h group,72 h group,120 h group,168 h group and 336 h group according to the sacrifice time,with 6 rats in each group.Rats in RFA group were treated with RFA to establish inflammatory model of liver injury;the rats in the control group were only connected with RFA equipment without power supply.The rats in the RFA group were sacrificed by decoupling at 12,24,72,120 and 168 h after RFA,and the rats in the control group were all sacrificed at 168 h.The serum levels of tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10) of rats in each group were measured by enzyme-linked immunosorbent assay(ELISA);the serum levels of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) of rats in each group were measured by 7170A automatic biochemical analyzer.The remaining six rats in the RFA group were sacrificed at 336 h,and the positive expression of M2 macrophages in hepatic tissue was detected by immunofluorescence method.Results The concentric circle-like liver injury inflammatory model of rats with a needle-centered necrotic area,a junction area,and an inflammatory hyperemia area was established successfully.With the development of time,the inflammation gradually lighten and the fibrosis repaired.The serum levels of ALT,AST,TNF-α and IL-10 of rats in the 12,24,72,120 and 168 h group were significantly higher than those in the control group(P<0.05).The serum levels of ALT,AST,TNF-α and IL-10 of rats in the 24,72 and 120 h group were significantly higher than those in the 12 h group(P<0.05).The serum level of AST of rats in the 168 h group was significantly higher than that in the 12 h group,but the serum level of TNF-α was significantly lower than that in the 12 h group(P<0.05);there was no significant difference in the serum ALT and IL-10 levels between the 168 h group and 12 h group(P>0.05).The serum levels of ALT,AST,TNF-α and IL-10 of rats in the 168 h group were significantly lower than those in the 120 h group(P<0.05).There was no significant difference in the serum levels of ALT,AST,TNF-α and IL-10 of rats among the 24,72 and 120 h group(P>0.05).The positive expression rates of M2 macrophages in the liver tissues of rats in the control group,12,24,72,120,168 and 336 h group were (9.5±0.4)%,(10.1±0.8)%,(10.9±0.5)%,(11.1±1.5)%,(18.7±0.8)%,(24.5±1.8)% and (6.8±0.4)%respectively.The positive expression rate of M2 macrophages in liver tissues of rats in 168 h group was significantly higher than that in control group,12,24,72,120 and 336 h group (χ2=14.91,14.31,13.60,13.36,12.71,17.63;P<0.05).Conclusion RFA can induce the polarization of liver M2 macrophages in rat,and its positive expression rate is the highest at 168 h after RFA.
