摘要:
目的 探讨基质金属蛋白酶-8(MMP-8)和MMP-9基因多态性与冠状动脉粥样硬化性心脏病(CAHD)患病风险的相关性。方法 选择2016年6月至2017年6月西安市第一医院收治的CAHD患者300例为CAHD组,另选择同期体检健康者300例为对照组,采集2组受试者血样,采用MassARRAY方法测定MMP-8和MMP-9基因上rs1940475、rs11225394、rs11225395、rs3918249、rs3918254和rs3787268位点的基因型,应用SPSS 19.0软件统计MMP-8和MMP-9基因多态性与CAHD患病风险的相关性。结果 MMP-8和MMP-9基因上6个候选单核苷酸多态性位点均符合Hardy-Weinberg平衡定律(P>0.05)。MMP-8基因上的rs11225394位点的等位基因T与降低CAHD患病风险有相关性[相对优势比(OR)=0.812,95%可信区间(CI):0.699~0.932,P=0.033]。MMP-9基因上的rs3918249和rs3787268位点的等位基因T和A亦与降低CAHD患病风险有相关性(rs3918249:OR=0.734,95%CI:0.613~0.998,P=0.035;rs3787268:OR=0.824,95%CI:0.693~0.978,P=0.024)。在最佳模型显性模型下,rs11225394位点的C/T和T/T基因型与降低CAHD患病风险有相关性(OR=0.760,95%CI:0.600~0.970,P=0.025);rs3918249位点的T/C和T/T基因型与降低CAHD患病风险有相关性(OR=0.720,95%CI:0.570~0.910,P=0.005);rs3787268位点的A/G和A/A基因型亦与降低CAHD患病风险有相关性(OR=0.740,95%CI:0.590~0.940,P=0.014)。连锁不平衡分析发现,MMP-9基因上的rs3918249、rs3918254和rs3787268位点组成的CCA单体型与降低CAHD患病风险具有相关性(OR=0.640,95%CI:0.340~0.950,P=0.026)。结论 MMP-8基因上的rs11225394位点和MMP-9基因上的rs3918249和rs3787268位点可能是CAHD的保护性位点。MMP-9基因上rs3918249、rs3918254和rs3787268位点组成的CCA单体型与降低CAHD风险具有相关性。
Abstract:
Objective To investigate the correlation between matrix metalloproteinases(MMP)-8 and MMP-9 gene polymorphism and the risk of coronary atherosclerotic heart disease (CAHD).Methods A total of 300 patients with CAHD in Xi′an No.1 Hospital from June 2016 to June 2017were selected as CAHD group,and 300 healthy persons were selected as control group in the same period.The blood samples of the subjects in the two groups were collected.The genotypes of rs1940475,rs11225394,rs11225395,rs3918249,rs3918254 and rs3787268 in MMP-8 and MMP-9 gene were detected by MassARRAY method;and the correlation between MMP-8 and MMP-9 gene polymorphism and the risk of CAHD was evaluated by SPSS 19.0 software.Results The six candidate single nucleotide polymorphisms loci in MMP-8 and MMP-9 genes were in line with the Hardy-Weinberg balance law (P>0.05).The allele T of rs11225394 locus in MMP-8 gene was correlated with reducing the risk of CAHD [odds ration(OR)=0.812,95%confidence intervals(CI):0.699-0.932,P=0.033].The allele T and A of rs3918249 and rs3787268 loci in MMP-9 gene were also associated with reducing the risk of CAHD (rs3918249:OR=0.734,95%CI:0.613-0.998,P=0.035;rs3787268:OR=0.824,95%CI:0.693-0.978,P=0.024).Under the best dominant model,the C/T and T/T genotypes of rs11225394 loci were correlated with reducing the risk of CAHD (OR=0.760,95%CI:0.600-0.970,P=0.025),the T/C and T/T genotype of rs3918249 locus were associated with reducing the risk of CAHD (OR=0.720,95%CI:0.570-0.910,P=0.005),the A/G and A/A genotypes of rs3787268 locus were also correlated with reducing the risk of CAHD (OR=0.740,95%CI:0.590-0.940,P=0.014).The linkage disequilibrium analysis showed that the CCA haplotype of rs3918249,rs3918254 and rs3787268 in MMP-9 gene was correlated with reducing the risk of CAHD (OR=0.640,95%CI:0.340-0.950,P=0.026).Conclusion The rs11225394 in MMP-8 gene and the rs3918249 and rs3787268 in MMP-9 gene may be the protective loci of CAHD.The CCA haplotype of rs3918249,rs3918254 and rs3787268 in MMP-9 gene are correlated with reducing the risk of CAHD.
