摘要:
目的 探讨免疫性血小板减少症(ITP)患者外周血中DNA甲基转移酶(DNMT)蛋白的表达水平及其与ITP发病之间的关系。方法 选择2015年5月至2016年5月新乡医学院第一附属医院收治的新诊断的急性ITP患儿60例作为观察组,另选择36例体检健康儿童作为对照组。采用酶联免疫吸附试验检测2组受试者血清DNMT1、DNMT3a及DNMT3b蛋白表达水平;比较观察组不同年龄和不同性别患儿血清DNMT1、DNMT3a及DNMT3b蛋白表达水平的差异;采用Pearson相关对观察组患儿血清DNMT1、DNMT3a及DNMT3b蛋白表达水平进行相关性分析。结果 观察组患儿血清DNMT1、DNMT3a及DNMT3b蛋白的表达水平低于对照组(P<0.05);不同年龄和不同性别患儿DNMT1、DNMT3a及DNMT3b蛋白表达水平比较差异均无统计学意义(P>0.05)。Pearson相关分析结果显示,观察组患儿血清DNMT1与DNMT3a、DNMT3b蛋白表达水平无相关性(r=-0.025、-0.057,P>0.05);DNMT3a与DNMT3b蛋白表达水平呈正相关(r=0.259,P<0.05)。结论 新诊断ITP患儿外周血中DNMT1、DNMT3a及DNMT3b蛋白表达水平显著降低,提示DNMT的异常表达可能与儿童ITP的发病相关。
Abstract:
Objective To investigate the expression of DNA methyltransferase (DNMT) protein in peripheral blood of patients with immune thrombocytopenia (ITP) and its relationship with the pathogenesis of ITP.Methods Sixty patients with newly diagnosed acute ITP in the First Affiliated Hospital of Xinxiang Medical University from May 2015 to May 2016 were selected as the observation group,and 36 healthy children were selected as the control group.The expressions of DNMTl,DNMT3a and DNMT3b protein in serum of subjects in the two groups were detected by enzyme linked immunosorbent assay method;the expressions of DNMT1,DNMT3a and DNMT3b protein in serum of children with different age and gender were compared in observation group;the correlation of DNMT1,DNMT3a and DNMT3b protein expression in the serum was analysed by Pearson correlation in the observation group.Results The expression levels of DNMT1,DNMT3a and DNMT3b protein in the serum of patients in observation group were significantly lower than those in the control group (P<0.05).There was no significant difference in the expression of DNMT1,DNMT3a and DNMT3b protein in children with different ages and sexes in observation group (P>0.05).Pearson correlation analysis showed that in the observation group there was no correlation between the expression of DNMT1 and DNMT3a and DNMT3b protein(r=-0.025,-0.057;P>0.05),and there was positive correlation between the expression of DNMT3a and DNMT3b protein(r=0.259,P<0.05).Conclusion The expression levels of DNMT1,DNMT3a and DNMT3b protein in serum of newly diagnosed ITP children are significantly decreased,suggesting that the abnormal expression of DNMT may be related to the pathogen esis of childhood ITP.
