摘要:
目的 探讨长链非编码RNA(lncRNA)HOXA11-AS在头颈部鳞状细胞癌组织中的表达及其对患者预后和癌细胞迁移的影响。方法 利用The Cancer Genome Atlas(TCGA)可视化数据库Gene Expression Profiling Interactive Analyses(GEPIA)分析lncRNA HOXA11-AS在头颈部鳞状细胞癌中的表达及其与患者预后的相关性。同时收集中南大学湘雅二医院口腔颌面外科2016年10月至2017年7月手术切除的24例头颈部鳞状细胞癌患者的癌组织和癌旁0.5 cm外正常组织,利用实时荧光定量聚合酶链反应(qRT-PCR)检测癌组织和癌旁组织中HOXA11-AS表达。取Tca8113细胞进行培养,待细胞生长至60%~70%融合时分为2组,实验组加HOXA11-AS siRNA进行转染,对照组加阴性对照siRNA进行转染,然后采用qRT-PCR法检测 2组细胞中HOXA11-AS的表达;采用Transwell细胞迁移实验检测2组细胞的迁移能力。结果 数据库分析结果显示,HOXA11-AS在头颈部鳞状细胞癌组织中的表达高于正常组织(P<0.01);HOXA11-AS表达与患者的总体生存率和无病生存率相关(HR=1.5、1.6,P=0.043 0、0.039 0)。24例头颈部鳞状细胞癌组织和癌旁组织qRT-PCR检测结果显示,HOXA11-AS在头颈部鳞状细胞癌组织和癌旁组织中的表达水平分别为2.58±0.37、0.95±0.29,HOXA11-AS在头颈部鳞状细胞癌组织中的表达明显高于癌旁组织(P<0.05)。细胞实验结果显示,实验组和对照组Tca8113细胞中HOXA11-AS表达分别为0.43±0.05、1.08± 0.07;对照组和实验组穿过Transwell小室的细胞数目分别为212±15、113±9个。实验组Tca8113细胞中HOXA11-AS表达低于对照组(P<0.05)。实验组穿过Transwell小室的细胞数目明显少于对照组(P<0.05)。结论 HOXA11-AS在头颈部鳞状细胞癌组织中高表达,且其能促进头颈部鳞状细胞癌细胞的迁移,有望成为头颈部鳞状细胞癌治疗的新靶点。
Abstract:
Objective To investigate the long non-coding RNA (lncRNA) HOXA11-AS expression in head and neck squamous cell carcinoma (HNSC) and its effect on the cell migration and prognosis of HNSC patients.Methods The expression of lncRNA HOXA11-AS in HNSC and the correlation of HOXA11-AS expression with prognosis of patients was analyzed by The Cancer Genome Atlas (TCGA)visualization database Gene Expression Profiling Interactive Analyses(GEPIA).The cancer tissue and normal tissues of 0.5 cm outside the carcinoma of 24 HNSC patients in the Department of Oral and Maxillofacial Surgery,the Second Xiangya Hospital of Central South University from October 2016 to March 2017 were collected.The expression of HOXA11-AS in the cancer tissue and paracancerous tissue was detected by real-time quantitative polymerase chain reaction(qRT-PCR).Tca8113 cells were cultured.When the cells grew to 60% to 70% fusion,they were divided into two groups.The cells in experimental group was transfected with HOXA11-AS siRNA;the cell in control group was transfected with negative control siRNA;then the expression of HOXA11-AS in the two groups was detected by qRT-PCR;the migration ability of Tca8113 cells in the two groups was detected by transwell chamber assay.Results Database analysis results showed that the expression of HOXA11-AS in HNSC tissues was significantly higher than that in normal tissues (P<0.01),and the high expression of HOXA11-AS was associated with the overall survival rate and disease-free survival rate of patients (HR=1.5,1.6;P=0.043 0,0.039 0).The qRT-PCR result of 24 HNSC patients showed that the expression of HOXA11-AS in cancer tissue and paracancerous tissue was 2.58±0.37、0.95±0.29 respectively;the expression of HOXA11-AS in cancer tissue was significantly higher than that in the paracancerous tissue (P<0.05).Cell experiment results showed that the expression of HOXA11-AS in Tca8113 cells in experimental group and control group was 0.43±0.05,1.08± 0.07 respectively;the number of cells passing through the Transwell chamber in the control group and experimental group was 212±15,113±9,respectively.The expression of HOXA11-AS in Tca8113 cells in experimental group was significantly lower than that in the control group(P<0.05);the number of cells passing through the Transwell chamber in the experimental group was significantly less than that in the control group (P<0.05).Conclusion The expression HOXA11-AS in HNSC is higher.And it can promote the migration of HNSC cells,which is expected to be a new target for the treatment of HNSC.
