摘要:
目的 探讨OBFC1基因多态性与脑胶质瘤患病风险的相关性。方法 选择2012年1月至2016年12月铜川矿务局中心医院收治的脑胶质瘤患者200例为脑胶质瘤组,另选择同期体检健康者200例为对照组,采集所有受试者血样,采用MassARRAY方法对OBFC1基因rs9325507、rs3814220、rs12765878和rs11191865等4个位点的基因型进行测定,分析OBFC1基因多态性与脑胶质瘤患病风险的相关性。结果 OBFC1基因上的rs9325507位点的等位基因“T”、rs3814220位点的等位基因“G”、rs12765878位点的等位基因“C”和rs11191865位点的等位基因“A”均会显著降低脑胶质瘤的患病风险(P<0.05)。在最佳模型显性模型下,rs9325507位点携带T/C和T/T基因型的个体脑胶质瘤患病风险显著降低,是携带C/C基因型的0.62倍(OR=0.620,95%可信区间为0.430~0.900,P=0.012);rs3814220位点携带G/A和G/G基因型的个体脑胶质瘤患病风险显著降低,是携带A/A基因型的0.62倍(OR=0.620,95%可信区间为0.430~0.910,P=0.014);rs12765878位点携带C/T和C/C基因型的个体脑胶质瘤患病风险显著降低,是携带T/T基因型的0.62倍(OR=0.620,95%可信区间为0.430~0.900,P=0.012);rs11191865位点携带A/G和A/A基因型的个体脑胶质瘤患病风险显著降低,是携带G/G基因型的0.65倍(OR=0.650,95%可信区间为0.450~0.940,P=0.023)。单体型分析结果显示,TGCA单体型会显著降低脑胶质瘤的患病风险(OR=0.760,95%可信区间为0.580~0.990,P=0.044)。结论 OBFC1基因上rs9325507、rs3814220、rs12765878和rs11191865等4个位点与降低脑胶质瘤的患病风险具有相关性。
Abstract:
Objective To investigate the correlation between the single nucleotide polymorphism (SNP) of OBFC1 and the risk of glioma.Methods A total of 200 patients with glioma treated in the Central Hospital of Tongchuan Mining Bureau from January 2012 to December 2016 were selected as glioma group,and 200 healthy subjects were selected as the control group in the same period.The blood samples were collected from all subjects.The genotype of rs9325507,rs3814220,rs12765878 and rs11191865 in OBFC1 gene was detected by MassARRAY method,and the correlation between OBFC1 gene polymorphism and the risk of glioma was analyzed.Results The allele "T" of rs9325507,the allele "G " of rs3814220,the allele "C" of rs12765878 and the allele "A" of rs11191865 locus could significantly reduce the risk of glioma (P<0.05).Under the best dominant model,the risk of glioma in the individual carrying the T/C and T/T genotypes at the rs9325507 locus was significantly reduced,it was 0.62 times as much as the individual carrying the C/C genotype(OR=0.620,95%CI:0.430-0.900,P=0.012).The risk of glioma in the individual carrying the G/A and G/G genotypes at rs3814220 locus was significantly lower,it was 0.62 times as much as the individual carrying the A/A genotype(OR=0.620,95%CI:0.430-0.910,P=0.014).The risk of glioma in the individual carrying the C/T and C/C genotypes at rs12765878 locus was significantly lower,it was 0.62 times as much as the individual carrying the T/T genotype(OR=0.620,95%CI:0.430-0.900,P=0.012).The risk of glioma in the individual carrying the A/G and A/A genotypes at rs11191865 locus was significantly lower,it was 0.65 times as much as the individual carrying the G/G genotype(OR=0.650,95%CI:0.450-0.940,P=0.023).Haplotype analysis showed that TGCA haplotype could significantly reduce the risk of glioma(OR=0.760,95%CI:0.580-0.990,P=0.044).Conclusion The rs9325507,rs3814220,rs12765878,and rs1119186 in OBFC1 gene are associated with reducing the risk of glioma.
