摘要:
目的 探讨双环醇对肾间质纤维化(RIF)大鼠肾组织中Caspase-3蛋白表达的影响及其对肾脏保护作用的可能机制。方法 选择13周龄健康雄性Sprague-Dawley大鼠90只,体质量200~250 g,随机分成假手术组、模型组和双环醇组,每组30只。模型组和双环醇组大鼠采用单侧输尿管梗阻法制作RIF模型,假手术组大鼠仅分离左侧输尿管。造模后,双环醇组大鼠立即给予双环醇灌胃(200 mg·kg-1),假手术组和模型组大鼠给予等量生理盐水灌胃,每日1次,连续21 d。光学显微镜观察大鼠肾脏外观及形态;苏木精-伊红染色和Masson染色观察肾间质病理学改变,并对肾小管间质损伤及RIF相对面积进行评价;免疫组织化学法检测各组大鼠肾组织中Caspase-3蛋白表达。结果 假手术组大鼠术后左肾形态无明显改变;模型组和双环醇组大鼠左肾肿胀,肾实质变薄,皮髓质界限模糊不清,肾盂及肾盏明显扩张。假手术组大鼠造模后第7、14、21天肾小管间质损伤程度、RIF相对面积及肾组织中Caspase-3蛋白表达水平两两比较差异无统计学意义(P>0.05);模型组和双环醇组大鼠造模后第14、21天肾小管间质损伤程度、RIF相对面积、肾组织中Caspase-3蛋白表达高于造模后第7天(P<0.05),造模后第21天肾小管间质损伤程度、RIF相对面积、肾组织中Caspase-3蛋白表达高于造模后第14天(P<0.05)。模型组和双环醇组大鼠造模后第7、14、21天肾小管间质损伤程度、RIF相对面积、肾组织中Caspase-3蛋白表达水平均高于假手术组(P<0.05);双环醇组大鼠术后第7、14、21天肾小管间质损伤程度、RIF相对面积、肾组织中Caspase-3蛋白表达水平均低于模型组(P<0.05)。大鼠肾组织中Caspase-3蛋白表达与肾小管间质损伤程度、RIF相对面积呈显著正相关(r=0.915、0.889,P<0.01)。结论 双环醇可能通过抑制RIF大鼠肾组织中Caspase-3蛋白表达而对肾组织起保护作用。
Abstract:
Objective To investigate the effect of bicyclol on expression of Caspase-3 protein in rat with renal interstitial fibrosis(RIF),and to explore protective mechanism of it on the nephridial tissue.Methods Ninty healthy Sprague-Dawley male rats,aged 13 weeks,weighted 200-250 g,were randomly divided into sham-operated group,model group and bicyclol group,with 30 rats in each group.RIF model of rats in the model group and the bicyclol group were established via unilateral ureteral obstruction.In the sham-operated group,left ureters were separated,but it weren′t obstructed and the rest procedures were the same as the other groups.After operations,the rats in the bicyclol group were given 200 mg·kg-1 bicyclol via intragastric administration once a day for 21 days,the rats in the sham-operated group and the model group were given identical voluminal normal saline via intragastric administrated.The histological examination of kidney tissue of rats were observed by light microscope.The pathology of renal interstitium changes were observed by routine hematoxylin and eosin and Masson staining,and the degree of mesenchyme of kidney tubules injury and relative area of RIF was estimated.The expression of Caspase-3 protein in renal tissue were determined with immunohistochemical method.Results There was no significant difference in the degree of mesenchyme of kidney tubules injury,relative area of RIF and the expression of Caspase-3 protein in nephridial tissue among the time points of 7,14,21 days after modeling in the sham-operated group.The degree of mesenchyme of kidney tubules injury,relative area of RIF and the expression of Caspase-3 protein in nephridial tissue at the time points of 14,21 days after modeling were higher than those at 7 days after modeling in the model group and the bicyclol group(P<0.05).The degree of mesenchyme of kidney tubules injury,relative area of RIF and the expression of Caspase-3 protein in nephridial tissue at 21 days after modeling were higher than those at 14 days after modeling in the model group and the bicyclol group(P<0.05).The degree of mesenchyme of kidney tubules injury,relative area of RIF and the expression of Caspase-3 protein in nephridial tissue in the model group and the bicyclol group at 7,14,21 days after modeling were higher than those in the sham-operated group(P<0.05).The degree of mesenchyme of kidney tubules injury,relative area of RIF and the expression of Caspase-3 protein in nephridial tissue in the bicyclol group at 7,14,21 days after modeling were lower than those in the model group(P<0.05).The expression of Caspase-3 protein in nephridial tissue was positive related with the degree of mesenchyme of kidney tubules injury and relative area of RIF(r=0.915,0.889;P<0.01).Conclusion Bicyclol has the predominant improve effect to the rats with RIF,the mechanism maybe involve in inhibiting expression of Caspase-3 protein in nephridial tissue.
