目的 探讨多药耐药相关蛋白P-糖蛋白(P-gp)、肺耐药蛋白(LRP)和谷胱甘肽巯基转移酶-π(GST-π)在原发性肝癌(HCC)患者外周血淋巴细胞及癌组织中的表达。方法 选择2014年1月至2015年1月在新乡医学院第一附属医院经影像学检查及病理学确诊的HCC患者69例(HCC组),采用免疫组织化学染色检测3种耐药相关蛋白P-gp、LRP和GST-π在HCC患者外周血淋巴细胞及癌组织中的表达水平;另选择40例健康人作为对照组,检测外周血淋巴细胞中P-gp、LRP和GST-π表达水平。结果 P-gp、LRP及GST-π在HCC组患者外周血淋巴细胞中的阳性表达率均高于对照组(P<0.05)。P-gp、LRP及GST-π在外周血淋巴细胞和癌组织中的阳性表达率比较差异均无统计学意义(P>0.05)。不同TNM分期HCC患者外周血淋巴细胞与癌组织中P-gp、LRP及GST-π阳性表达率比较差异均无统计学意义(P>0.05)。结论 多药耐药相关蛋白P-gp、LRP和GST-π在HCC患者外周血淋巴细胞及癌组织中的表达一致,可以使用外周血淋巴细胞检测多药耐药相关蛋白。
Abstract:
Objective To investigate the expression of multidrug resistance related proteins of P-glycoprotein (P-gp),lung resistance protein (LRP),and glutathione S-transferase-π(GST-π) in peripheral blood lymphocytes and cancer tissues in patients with hepatocellular carcinoma(HCC).Methods Sixty-nine HCC patients who were diagnosed by imaging detection and pathology in the First Affiliated Hospital of Xinxiang Medical University from January 2014 to January 2015 were selected.The expression levels of P-gp,LRP,and GST-π in peripheral blood lymphocytes and cancer tissues were detected by immunohistochemical staining method.Forty healthy people were chosen as control group,and the expression levels of P-gp,LRP and GST-π in peripheral blood lymphocytes were detected.Results The positive expression rates of P-gp,LRP and GST-π in peripheral blood lymphocytes of patients in the HCC group were significantly higher than those in the control group(P<0.05).There was no significant difference in the positive expression rates of P-gp,LRP and GST-π between peripheral blood lymphocytes and cancer tissues of patients in HCC group(P>0.05).There was no significant difference in positive expression rates of P-gp,LRP and GST-π between peripheral blood lymphocytes and cancer tissues of HCC patients with different TNM stage(P>0.05).Conclusion The expression of multidrug resistance related protein of P-gp,LRP and GST-π in peripheral blood lymphocytes and cancer tissues in patients with HCC is consistent.The peripheral blood lymphocytes can be used to detect the expression of multidrug resistance related protein.
