摘要:
目的 探讨白藜芦醇对急性心肌梗死(AMI)大鼠核因子-κB(NF-κB)信号通路表达的干预作用。方法 将30只雄性Sprague-Dawley大鼠随机分为假手术组、AMI组和白藜芦醇组,每组10只;AMI和白藜芦醇组大鼠采用结扎大鼠左冠状动脉前降支法制备AMI模型,假手术组大鼠不结扎左冠状动脉前降支;术后第1天白藜芦醇组大鼠开始腹腔注射白藜芦醇10 mg·kg-1·d-1,假手术组及AMI组大鼠分别注射等剂量生理盐水;每日 1次,持续用药至术后4周。然后处死各组大鼠,取梗死区域的心肌组织,采用实时荧光定量聚合酶链反应和Western blot法检测大鼠心肌组织中NF-κB、NF-κB抑制蛋白α(IκBα)、IκB激酶α(IKKα)蛋白及其mRNA的表达。结果 AMI组大鼠心肌组织中NF-κB、IκBα和IKKα mRNA表达显著高于假手术组(P<0.05),白藜芦醇组大鼠心肌组织中NF-κB、IκBα和IKKα mRNA表达显著低于AMI组(P<0.05),白藜芦醇组与假手术组大鼠心肌组织中NF-κB、IκBα和IKKα mRNA表达比较差异均无统计学意义(P>0.05)。AMI组大鼠心肌组织中NF-κB、IκBα和IKKα蛋白表达显著高于假手术组(P<0.05),白藜芦醇组大鼠心肌组织中NF-κB、IκBα和IKKα蛋白表达显著低于AMI组(P<0.05);白藜芦醇组大鼠心肌组织中NF-κB、IκBα蛋白表达显著高于假手术组(P<0.05),但2组大鼠心肌组织中IKKα蛋白表达比较差异无统计学意义(P>0.05)。结论 白藜芦醇可以抑制AMI大鼠NF-κB信号通路表达情况,具有心脏保护效应。
Abstract:
Objective To investigate the intervention effect of resveratrol on nuclear factor-κB(NF-κB) signal pathway in rats with acute myocardial infarction(AMI).Methods Thirty male Sprague-Dawley rats were randomly divided into sham operation group,AMI group and resveratrol group,ten rats in each group.The AMI rat models were established by ligating the left anterior descending coronary artery in AMI group and resveratrol group,but the rats in sham operation group were not ligated the left anterior descending coronary artery.On the first day after operation,the rats in the resveratrol group received resveratrol 10 mg·kg-1·d-1 by intraperitoneal injection,and the rats in sham operation group and AMI group received isodose saline by intraperitoneal injection,once a day for four weeks.Then the rats in each group were sacrificed and the myocardial tissues of infarction area were collected.The expressions of NF-κB,NF-κB inhibitory protein-α(IκBα),IκB kinase-α(IKKα) protein and mRNA in myocardial tissues of rats were detected by real time quantitative polymerase chain reaction and Western blot method.Results The expressions of NF-κB,IκBα and IKKα mRNA in myocardial tissues of rats in AMI group were significantly higher than those in sham operation group(P<0.05).The expressions of NF-κB,IκBα and IKKα mRNA in myocardial tissues of rats in resveratrol group were significantly lower than those in AMI group(P<0.05).There was no significant difference in the expressions of NF-κB,IκBα and IKKα mRNA in myocardial tissues of rats between resveratrol group and sham operation group(P>0.05).The expressions of NF-κB,IκBα and IKKα protein in myocardial tissues of rats in AMI group were significantly higher than those in sham operation group(P<0.05).The expressions of NF-κB,IκBα and IKKα protein in myocardial tissues of rats in resveratrol group were significantly lower than those in AMI group(P<0.05).The expressions of NF-κB and IκBα protein in myocardial tissues of rats in resveratrol group were significantly higher than those in sham operation group(P<0.05),but there was no significant difference in the expression of IKKα protein in myocardial tissues of rats between resveratrol group and sham operation group(P>0.05).Conclusion Resveratrol can protect the heart by inhibiting the expression of NF-κB signal pathway in AMI rats.
