摘要:
目的 探讨蛋白激酶C(PKC)α、βⅡ、ε、ζ在结肠腺癌中的表达及与结肠腺癌发生、发展及转移的关系。方法 选择2010年4 月至2012年8月大庆油田总医院收治的82例原发性结肠腺癌患者的手术切除标本,其中管状腺癌52例,黏液腺癌30例;淋巴结转移39例,无淋巴结转移43例。采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶法检测癌组织标本中PKCα、βⅡ、ε、ζ蛋白的表达。结果 52例结肠管状腺癌中,PKCα、βⅡ、ζ 3个亚型的阳性表达率在高、中、低各分化组之间比较差异均无统计学意义(P>0.05);而PKCε亚型的阳性表达率在高、中分化组之间以及中、低分化组之间比较差异均无统计学意义(P>0.05),但高分化组的阳性表达率显著低于低分化组(χ2=31.523,P<0.05)。黏液腺癌组织中PKCα、βⅡ的阳性表达率显著低于管状腺癌组织(P<0.05),而PKCε的阳性表达率则显著高于管状腺癌组织(P<0.05);黏液腺癌组织与管状腺癌癌组织中PKCζ的阳性表达率比较差异无统计学意义(P>0.05)。原发性结肠腺癌患者中无淋巴结转移者癌组织中PKCα的阳性表达率显著高于有淋巴结转移者(P<0.05);无淋巴结转移者癌组织中PKCβⅡ、ε、ζ 的阳性表达率与有淋巴结转移者比较差异无统计学意义(P>0.05)。结论 PKC各亚型在结肠腺癌发生、发展和转移中的作用不同,PKCε在结肠管状腺癌中的表达与分化有关,PKCε可能参与了黏液腺癌的黏液分泌,PKCα、βⅡ可能与结肠管状腺癌的细胞增殖有关;PKCα与结肠腺癌淋巴结转移有关。
Abstract:
Objective To investigate the relationship between the expressions of protein kinase C (PKC)α,PKCβⅡ,PKCε,PKCζ and the occurrence,development,metastasis of colon adenocarcinoma.Methods A total of 82 specimens of primary colon carcinoma were selected from April 2010 to August 2012 in Daqing Oilfield General Hospital.Among the specimens,there were 52 cases of tubular adenocarcinoma and 30 cases of mucinous adenocarcinoma.There were 39 cases with lymph node metastasis and 43 cases without lymph node metastasis.The expressions of PKCα,PKCβⅡ,PKCε and PKCζ in colon carcinoma tissue were detected by streptavidin peroxidase method of immunohistochemistry.Results Among the 52 cases of tubular adenocarcinoma of colon,there was no significant difference in the positive expression rates of PKCα,PKCβⅡ and PKCζ among the well,middle and poor differentiation tubular adenocarcinoma (P>0.05).There was no significant difference in the positive expression rate of PKCε between the well differentiation group and middle differentiation group and between the middle differentiation group and poor differentiation group (P>0.05);but the positive expression rate of PKCε in well differentiation group was significantly lower than that in poor differentiation group(χ2=31.523,P<0.05).The positive expression rates of PKCα and PKCβⅡ in mucinous adenocarcinoma tissue were significantly lower than those in tubular adenocarcinoma (P<0.05),the positive expression rate of PKCε in mucinous adenocarcinoma was significantly higher than that in tubular adenocarcinoma (P<0.05),there was no significant difference in the positive expression rate of PKCε between mucinous adenocarcinoma and tubular adenocarcinoma (P>0.05).The positive expression rate of PKCα in colon adenocarcinoma without lymph node metastasis was significantly higher than that in colon adenocarcinoma with lymph node metastasis(P<0.05).There was no significant difference in the positive expression rates of PKCβⅡ,PKCε and PKCζ between colon adenocarcinoma with lymph node metastasis and colon adenocarcinoma without lymph node metastasis(P<0.05).Conclusion PKC subtypes play the different roles in the occurrence,development and metastasis of colon adenocarcinoma.The expression of PKC is related to the differentiation of tubular adenocarcinoma,and PKC may be involved in the mucus secretion of mucinous adenocarcinoma.PKCα and PKCβⅡ may be related to the cell multiplication of tubular adenocarcinoma,and PKCα is associated with lymph node metastasis in colon adenocarcinoma.
