摘要:
目的 观察心脑宁对D-半乳糖诱导的皮肤衰老小鼠的干预性作用。方法 将40只雌性小鼠随机分为模型组、心脑宁组、生理盐水组和对照组,每组10只。对照组小鼠不进行任何处理;其余3组小鼠于颈背部皮下注射D-半乳糖125 mg·kg-1·d-1,连续42 d。在注射D-半乳糖21 d后,模型组不给予任何干预措施,心脑宁组小鼠给予心脑宁灌胃(0.375 g·kg-1·d-1),生理盐水组小鼠给予等量生理盐水灌胃,连续灌胃21 d 后,处死所有小鼠。分别检测血清与皮肤组织中超氧化物歧化酶(SOD)活性及丙二醇(MDA)和羟脯氨酸(HYP)水平。结果 模型组小鼠血清与皮肤组织中SOD活性和HYP水平明显低于对照组,MDA水平明显高于对照组(P<0.01)。心脑宁组小鼠血清与皮肤组织中SOD活性和HYP水平明显高于模型组和生理盐水组(P<0.01);MDA水平明显低于模型组和生理盐水组(P<0.01)。模型组小鼠血清与皮肤组织中SOD活性及HYD、MDA水平与生理盐水组比较差异均无统计学意义(P>0.05)。心脑宁组小鼠血清和皮肤组织中SOD活性及MDA、HYP水平与对照组比较差异均无统计学意义(P>0.05)。结论 心脑宁可阻止由D-半乳糖引起的皮肤衰老现象,对延缓和预防皮肤衰老具有一定作用。
Abstract:
Objective To observe the intervention effect of Xinnaoning on skin aging mice induced by D-galactose.Methods Forty female mice were randomly divided into model group,Xinnaoning group,saline group and control group,with 10 mice in each group.The mice in control group were not given any intervention.Another mice were injected subcutancously D-galactose(125 mg·kg-1·d-1)once per day for 42 d.After 21 d when the mice were injected D-galactose,the mice in Xinnaoning group were given Xinnaoning by intragastric administration for 21 d;the mice in saline group were given the same volume saline by intragastric administration for 21 d;the mice in model group were not given any intervention.Then all mice were sacrificed and the activity of superoxide dismutase(SOD),the contents of malondialdehyde(MDA) and hydroxyproline(HYP) in serum and skin tissues were detected respectively.Results The activity of SOD,and the contents of HYP in serum and skin tissues of mice in model group were significantly lower than those in the control group;but the content of MDA was significanlty higher than that in the control group(P<0.01).The activity of SOD,and the contents of HYP in serum and skin tissues of mice in Xinnaoning group were significantly higher than those in the model group and saline group;but the content of MDA was significanlty lower than that in the model group and saline group (P<0.01).There was no statistic difference in the activity of SOD,and the contents of HYP and MDA in serum and skin tissues of mice between the model group and saline group(P>0.05).There was no statistic difference in the activity of SOD,and the contents of HYP and MDA in serum and skin tissues of mice between the Xinnaoning group and control group(P>0.05).Conclusion Xinnaoning can prevent the skin aging caused by D-galactose and has certain role in delaying and preventing skin aging.
