摘要:
目的 探讨大黄附子汤对大鼠功能性便秘的治疗作用。方法 成年Sprague Dawley雄性大鼠30只,随机分为空白对照组、模型组和大黄附子汤组,每组10只。空白对照组大鼠正常饲养,其余大鼠采用复方地芬诺酯(20 mg·kg-1)连续灌胃2周,建立大鼠功能性便秘模型。造模成功后模型组大鼠以生理盐水灌胃;大黄附子汤组大鼠以大黄附子汤2 mL灌胃,共给药1周;空白对照组大鼠以相同体积的生理盐水灌胃。观察各组大鼠体质量和排便量;墨汁灌胃法测定小肠推动力,Power Lab检测大鼠在体结肠肌电,苏木精-伊红染色观察结肠的病理变化。结果 与空白对照组相比,模型组大鼠体质量增加缓慢,大便量减少,小肠推动力减弱,结肠肌电慢波频率减慢,振幅减小(P<0.05,P<0.01);与模型组比较,大黄附子汤组大鼠体质量增加较快,大便量增加,小肠推动力增强,结肠肌电慢波频率加快,振幅增加(P<0.05,P<0.01)。空白对照组大鼠肠腺稠密,黏膜杯状细胞数目较多;模型组大鼠肠腺稀薄,黏膜杯状细胞数目减少;与模型组相比,大黄附子汤组大鼠肠腺相对稠密,黏膜杯状细胞数目增多。结论 大黄附子汤对大鼠功能性便秘具有治疗作用。
Abstract:
Objective To explore the therapeutical effect of Rhei and Aconiti Praeparatae Decoction on functional constipation rats.Methods Thirty adult male Sprague Dawley rats were randomly divided into blank control group,model group,and Rhei and Aconiti Praeparatae Decoction group,with 10 rats in each group.The rats in blank control group were fed normally;the other rats were given compound diphenoxylate tablets (20 mg·kg-1)for two weeks to establish the functional constipation model.After the success of modeling,the rats in model group were given physiological saline intragastric administration;the rats in Rhei and Aconiti Praeparatae Decoction group were given Rhei and Aconiti Praeparatae Decoction (2 mL) by intragastric administration for one week;the rats in blank control group were given the same volume of physiological saline.The body weight and defecation quantity of rats in each group were observed;the driving force of small intestine was measured by oral administration of ink;the colonic electromyography was detected by Power Lab;the pathological changes of colon were detected by haematoxylin-haematoxylin (HE) staining.Results Compared with the blank control group,the rats in model group showed reduced body weight increment,decreased defecation quantity,weakened small intestine driving force,decreased frequency and amplitude of in vivo colonic electromyography slow wave (P<0.05 or P<0.01).Compared with model group,the rats in Rhei and Aconiti Praeparatae Decoction group has increased body weight increment and defecation quantity,strengthened small intestine driving force,increased frequency and amplitude of in vivo colonic electromyography slow wave (P<0.05 or P<0.01).The rats in blank control group had dense intestinal gland and more goblet cells;the rats in model group had tenuity intestinal gland and less goblet cells;compared with model group,the rats in Rhei and Aconiti Praeparatae Decoction group had relatively dense intestinal gland and more goblet cells.Conclusion Rhei and Aconiti Praeparatae Decoction has optimal therapeutic efficacy on functional constipation rats.
