摘要:
目的 探讨核因子-κB(NF-κB)在体外培养离体的公猪主动脉瓣膜间质细胞(AVIC)钙化过程中表达的变化。方法 体外培养公猪AVIC,镜下观察其形态,应用免疫荧光染色法对AVIC进行鉴定。将AVIC分为对照组和实验组,对照组AVIC在标准培养基中培养2周,实验组AVIC在钙化诱导液中培养2周。光学显微镜下对钙化结节行茜素红染色,并进行计数;应用Western blot检测2组AVIC中NF-κB蛋白表达水平。结果 成功分离并体外培养了离体的公猪AVIC,免疫荧光结果显示,α-平滑肌肌动蛋白及波形蛋白染色阳性,血管性血友病因子染色阴性。实验组AVIC自发形成钙结节数及AVIC 中NF-κB表达水平均较对照组显著增高(P<0.05)。结论 钙化后公猪AVIC中NF-κB表达水平增高,NF-κB可能与主动脉瓣膜钙化的发生有关。
Abstract:
Objective To investigate the changes of nuclear factor-κB(NF-κB) in aortic valvular interstitial cells (AVICs) cultured in vitro from boar during the process of calcification.Methods AVICs were isolated and cultured,and the morphous was observed under microscope.Fluorescent staining was performed to identify the AVIC.The AVIC was divided into experimental group and control group:the AVICs in control group were training in the standard medium for 2 weeks,while the the cells in experimental group were inducing calcification for 2 weeks.Calcium deposit was stained by alizarin red and calcified nodules was quantified under microscope.Western blot was used to detect the expression level of NF-κB in the two groups.Results AVICs of boar were successfully harvested and identified in vitro.It showed that positive staining of α-smooth muscle actin,vimentin,and negative staining of von willebrand factor.The calcified nodules and the expression of NF-κB in the experimental group were higher significantly than those in the control group(P<0.05).Conclusion The increasing of the expression of NF-κB after inducing calcification may be associated with the occurrence of aortic valve calcification in some degree.
