摘要:
目的 探讨磷脂酸磷酸酶2域1A(PPAPDC1A)基因在正常乳腺上皮细胞和不同转移能力的乳腺癌细胞中表达的差异。方法 以正常乳腺上皮HBL-100细胞、低转移乳腺癌MCF-7细胞、高转移乳腺癌MDA-MB-435s细胞和高转移乳腺癌MDA-MB-231细胞为研究对象,通过实时荧光定量聚合酶链反应(qPCR)和Western blot法分别检测PPAPDC1A基因mRNA和蛋白在上述细胞中的表达。结果 与正常乳腺上皮HBL-100细胞比较,3种不同转移能力的乳腺癌细胞中的PPAPDC1A mRNA和蛋白表达水平均升高(P<0.01),且高转移乳腺癌MDA-MB-231细胞和高转移乳腺癌MDA-MB-435s细胞中PPAPDC1A mRNA和蛋白表达水平均高于低转移乳腺癌MCF-7细胞(P<0.01),高转移乳腺癌MDA-MB-231细胞中PPAPDC1A mRNA和蛋白水平的表达高于高转移乳腺癌MDA-MB-435s细胞(P<0.01)。PPAPDC1A在4种细胞中表达的顺序为HBL-100细胞<MCF-7细胞<MDA-MB-435s细胞<MDA-MB-231细胞。结论 PPAPDC1A基因可能促进了乳腺癌的发生、转移。
Abstract:
Objective To investigate the expression difference of phosphatidic acid phosphatase type 2 domain containing 1A(PPAPDC1A)gene in normal mammary epithelial cells and breast carcinoma cells with different metastatic abilities.Methods The mRNA and protein expressions of PPAPDC1A gene in normal mammary epithelial HBL-100 cells,low metastatic breast carcinoma MCF-7cells,high metastatic breast carcinoma MDA-MB-231 cells and MDA-MB-435s cells were detected by real-time quantitative PCR(qPCR) and Western blot assay,respectively.Results The qPCR and Western blot results showed that,compared with normal mammary epithelial HBL-100 cells,the mRNA and protein expressions of PPAPDC1A gene in three different metastatic ability of breast carcinoma cells were significantly increased(P<0.01).Furthermore,the mRNA and protein expressions of PPAPDC1A gene in high metastatic breast carcinoma MDA-MB-231 cells and MDA-MB-435s cells were significantly higher than those in the low metastatic breast carcinoma MCF-7 cells(P<0.01),and the mRNA and protein expressions of PPAPDC1A gene in high metastatic breast carcinoma MDA-MB-231 cells were significantly higher than those in the high metastatic breast carcinoma MDA-MB-435s cells(P<0.01).The expression order of PPAPDC1A gene in four kinds of cells was as follows:HBL-100 cells<MCF-7 cells<MDA-MB-435s cells<MDA-MB-231 cells.Conclusion The PPAPDC1A gene may promote the occurrence and metastasis of breast carcinoma.
